Abstract
Description
中空構造を有している培養容器と、前記培養容器内に設けられ、前記培養容器の内部空間を複数の区画へ区分する隔壁とを備えたデバイス本体であって、前記培養容器は、前記内部空間と外部空間とを各々が連通させる1以上の流入孔と流出孔とが設けられ、前記複数の区画の位置で光透過性を有している光透過部分を有し、前記隔壁は、前記複数の区画の各々と前記1以上の流入孔との間及び前記複数の区画の各々と前記流出孔との間での液体の移動を可能とするデバイス本体と、
前記複数の区画の各々を満たした培養液と、
前記内部空間内に位置した複数種類の微生物と
を備え、
前記複数の区画の少なくとも1つには、前記複数種類の微生物の2種類以上が存在し且つ前記複数種類の微生物の1種類以上が存在しない微生物観察用デバイスが提供される。
内部空間と、前記内部空間と外部空間とを各々が連通させる1以上の流入孔と流出孔とが設けられ、光透過性を有している光透過部分を有している培養容器を準備することと、
複数種類の微生物と光硬化性樹脂と培養液とを含んだ微生物含有液を前記内部空間に供給することと、
前記光透過部分を介して前記微生物含有液の一部に光を照射して、前記光硬化性樹脂の硬化物からなり、前記内部空間を複数の区画に区分する隔壁を、前記複数の区画の各々と前記1以上の流入孔との間、及び前記複数の区画の各々と前記流出孔との間での液体の移動を可能とし、且つ前記複数の区画の少なくとも1つに、前記複数種類の微生物の2種類以上が存在し且つ前記複数種類の微生物の1種類以上が存在しないように形成することと
を含んだ微生物観察用デバイスの製造方法が提供される。
図1は、本発明の実施形態に係る微生物観察用デバイスの上面図の一例を示す模式図である。
この微生物観察用デバイス1は、デバイス本体2と培養液3と複数種類の微生物4とを備えている。
図2に示すように、培養容器20は、第1基板20Aと、第1基板20Aと貼り合わされた板状の第2基板20Bとからなる。第2基板20Bと対向している側の、第1基板20Aの主面には、2つの貫通孔と、これら貫通孔を連通させる凹部とが設けられている。2つの貫通孔及び凹部は、それぞれ、流入孔200、流出孔201及び内部空間202に対応している。
図3に示すD3は、第3部分A3の横幅である。横幅D3は、100μm乃至10000μmの範囲内にあることが好ましい。
また、各区画Pの縦幅は、10μm乃至100μmの範囲内にあることが好ましい。
図1に示す微生物観察用デバイス1は、例えば、以下の方法により製造することができる。
以上、図1に示す微生物観察用デバイス1の製造方法について述べた。
上述した微生物観察用デバイス1は、一例によると、微生物の培養方法に用いることができる。微生物の培養方法は、例えば、上述した1以上の流入孔200を介して、培養液を連続又は断続的に供給して複数種類の微生物4を培養することを含む。培養液を連続又は断続的に供給する場合、培養液の組成は経時的に変更させてもよい。
上述した微生物の培養及び観察は同時に行うことも可能である。
図5は、他の実施形態に係る微生物観察用デバイス1の上面図の一例を示す模式図である。図5に示す微生物観察用デバイス1では、培養容器20に2つの流入孔200、即ち、流入孔200Aと流入孔200Bとが設けられている。また、図5に示すデバイス本体2の第1部分A1、即ち、内部空間202内であって、流入孔200A及び流入孔200Bと複数の区画Pからなる配列との間に、濃度勾配生成部Gが設けられている。図5に示す微生物観察用デバイス1は、培養容器20に2つの流入孔が設けられていることと、デバイス本体2に濃度勾配生成部Gが設けられていること以外は、図1に示す微生物観察用デバイス1と同じである。
第1濃度勾配生成部G1は、各々が分岐した2つの第1流路F1と3つの第2流路F2とを含んでいる。2つ第1流路F1は、それらの入口が、流入孔200A及び流入孔200Bにそれぞれ接続されている。各第2流路F2は蛇行している。3つの第2流路F2の2つの各々は、その入口が2つの第1流路F1の1つが有している出口の1つに接続されている。また、3つの第2流路F2の残りは、その入口が2つの第1流路F1の2つの一方が有している出口の1つと他方が有している出口の1つとに接続されている。
以上、図5に示す微生物観察用デバイス1について述べた。
以上、図6に示す微生物観察用デバイス1について述べた。
以上、図7に示す微生物観察用デバイス1について述べた。
ところで、複数種類の微生物を共培養する方法の一例として、区画が設けられていない培養容器で複数種類の微生物を培養する方法がある。しかしながら、この方法によって培養した複数種類の微生物を観察する場合、培養容器から観察用プレートへ複数種類の微生物を移動させる必要があった。このため、このような方法では観察する際に手間が生じる。また、微生物の種類が多い場合においては、観察する際に特に手間が生じる。
先ず、培養液供給部70から、ドロップレット生成部71に微生物培養液700が断続的に供給される。次に、ドロップレット生成部71に供給された微生物培養液700がドロップレット生成部71を通過し、ドロップレット収容部73に入る際に、油膜材料供給部72によってドロップレット生成部71に供給された油膜材料によって微生物培養液700がカプセル化される。このカプセル化によってドロップレット74が生成される。

Claims (8)
- 中空構造を有している培養容器と、前記培養容器内に設けられ、前記培養容器の内部空間を複数の区画へ区分する隔壁とを備えたデバイス本体であって、前記培養容器は、前記内部空間と外部空間とを各々が連通させる1以上の流入孔と流出孔とが設けられ、前記複数の区画の位置で光透過性を有している光透過部分を有し、前記隔壁は、前記複数の区画の各々と前記1以上の流入孔との間及び前記複数の区画の各々と前記流出孔との間での液体の移動を可能とするデバイス本体と、
前記複数の区画の各々を満たした培養液と、
前記内部空間内に位置した複数種類の微生物と
を備え、
前記複数の区画の少なくとも1つには、前記複数種類の微生物の2種類以上が存在し且つ前記複数種類の微生物の1種類以上が存在しない微生物観察用デバイス。 - 前記培養容器の内面は、互いに向き合った第1領域及び第2領域を含み、前記隔壁は、前記第1領域から前記第2領域へ向けて伸び且つ前記第2領域から離間している請求項1に記載の微生物観察用デバイス。
- 前記1以上の流入孔は2以上の流入孔であり、
前記デバイス本体には、前記内部空間内であって、前記2以上の流入孔と前記複数の区画からなる配列との間に、各々が分岐した2以上の第1流路と3以上の第2流路とを含んだ濃度勾配生成部が設けられ、
前記2以上の第1流路はそれらの入口が前記2以上の流入孔にそれぞれ接続され、
前記3以上の第2流路の2つの各々は、その入口が前記2以上の第1流路の1つが有している出口の1つに接続され、
前記3以上の第2流路の残りの各々は、その入口が前記2以上の第1流路の2つの一方が有している出口の1つと他方が有している出口の1つとに接続された請求項1又は2に記載の微生物観察用デバイス。 - 前記複数の区画は、第1方向へ配列した複数の区画から各々がなる1以上の列を形成しており、前記1以上の流入孔と、前記1以上の列と、前記流出孔とは、前記第1方向に対して交差する第2方向へこの順に配列している請求項1乃至3の何れか1項に記載の微生物観察用デバイス。
- 内部空間と、前記内部空間と外部空間とを各々が連通させる1以上の流入孔と流出孔とが設けられ、光透過性を有している光透過部分を有している培養容器を準備することと、
複数種類の微生物と光硬化性樹脂と培養液とを含んだ微生物含有液を前記内部空間に供給することと、
前記光透過部分を介して前記微生物含有液の一部に光を照射して、前記光硬化性樹脂の硬化物からなり、前記内部空間を複数の区画に区分する隔壁を、前記複数の区画の各々と前記1以上の流入孔との間、及び前記複数の区画の各々と前記流出孔との間での液体の移動を可能とし、且つ前記複数の区画の少なくとも1つに、前記複数種類の微生物の2種類以上が存在し且つ前記複数種類の微生物の1種類以上が存在しないように形成することと
を含んだ微生物観察用デバイスの製造方法。 - 前記培養容器の内面は、互いに向き合った第1領域及び第2領域を含み、前記隔壁は、前記第1領域から前記第2領域へ向けて伸び且つ前記第2領域から離間するように形成する請求項5に記載の微生物観察用デバイスの製造方法。
- 請求項1乃至4の何れか1項に記載の微生物観察用デバイスに、前記1以上の流入孔を介して、前記培養液を連続又は断続的に供給して前記複数種類の微生物を培養することを含んだ微生物の培養方法。
- 請求項1乃至4の何れか1項に記載の微生物観察用デバイスが備えている前記複数種類の微生物のうち、前記複数の区画の1以上に位置しているものを、前記光透過部分を介して観察することを含んだ微生物の観察方法。
