本願は、新規がん抗原ペプチドに関する。
KILQQSRIVQX(ここで、Xは存在しないか、またはSである)のアミノ酸配列;
DVQKIVESQINFHGKKLKLGPAIRKQNLCAYHVQPRPL(配列番号16)のアミノ酸配列中の連続する10個以上のアミノ酸の配列;または、
QNLNHYIQVLENLVRSVPS(配列番号9)のアミノ酸配列
を含む、10個~45個のアミノ酸からなるペプチド、または、
前記のいずれかのペプチドにおいて1個~3個のアミノ酸が置換、欠失または付加され、かつ、ヘルパーT細胞活性化能を有するペプチドを提供する。

を含み、ヘルパーT細胞またはそれにより産生されるサイトカインの量の増加がステルスがん抗原特異的ヘルパーT細胞の存在または量を示すものである方法を提供する。試料は、抗原提示細胞を含むものであればいかなるものであってもよく、例えば、末梢血単核球、浸潤性リンパ球、腫瘍細胞、腹水中の細胞、胸水中の細胞、脳脊髄液中の細胞、骨髄細胞、またはリンパ節細胞などが挙げられる。試料は、健常人由来であっても、がん患者由来であってもよい。健常人由来の試料を用いることにより、例えば、がんに罹患しているかどうか、あるいはその素因を有するかどうかを診断することなどが可能になる。がん患者由来の試料を用いることにより、例えば、当該がん患者においてステルスがん抗原を用いる免疫療法が効果を有するかどうかを予測することなどが可能になる。ある実施形態において、SYCP3またはSPESP1に由来するヘルパーペプチド特異的ヘルパーT細胞の量を決定される対象は、HLA-DR陽性、特にはHLA-DR53陽性であるが、これに限定されない。ある実施形態において、DAZL1に由来するヘルパーペプチド特異的ヘルパーT細胞の量を決定される対象は、HLA-DR陽性、特にはHLA-DR4、8、9、15または53陽性であるが、これに限定されない。取得した試料は、ヘルパーペプチドによる刺激の前後に培養されてもよく、前記培養条件は当業者が適宜決定することができる。ヘルパーペプチドを用いたこれらの細胞の刺激は、公知の手法を用いて行うことができ、インビトロまたはインビボのいずれにおいて行われてもよい。ヘルパーT細胞またはそれにより産生されるサイトカインの量は、公知の方法により調べることができる。

上記の説明は、すべて非限定的なものであり、添付の特許請求の範囲において定義される本発明の範囲から逸脱せずに、変更することができる。さらに、下記の実施例は、すべて非限定的な実施例であり、本発明を説明するためだけに供されるものである。
DNAメチル基転移酵素阻害剤(5-アザ-2'-デオキシシチジン(デシタビン);5-AZA)によって遺伝子発現が再上昇する分子を同定するために、肺癌細胞株A549を5-AZA(10μM)にて3日間処理後にRNAを抽出し、DNAマイクロアレイにて未処理の癌細胞株において発現量が0に近く、かつ5-AZA処理癌細胞株における発現量が30倍以上増加している分子を探索した。その結果、SYCP3(未処理:3.0、5-AZA処理後:340.9、発現上昇率:112.3)、SPESP1(未処理:1.7、5-AZA処理後:65.3、発現上昇率:38.8)およびDAZL1(未処理2.8、5-AZA処理後:341.3、発現上昇率:123.6)を含む候補分子を同定した。
各種の癌細胞株(肺癌細胞株EBC1、肺癌細胞株Lu65、大腸癌細胞株HT-29、口腔扁平上皮癌細胞株SAS)を、5-AZA(10μM)を含む完全培地(100U/mLペニシリン(明治製菓)、100μg/Lストレプトマイシン(明治製菓)加RPMI1640培地(ナカライテスク 30264-56)に56℃、30分非働化した牛胎児血清(biosera FB-1365/500)を10%添加したもの)を用いて2x105/ウェルの条件で6穴培養プレート(Falcon 353046)にて、温度37℃、CO2濃度5%、湿度95%に設定したインキュベーター(SANYO)内で3日間培養した(以下、全ての細胞培養はこのインキュベーターを用いた)。2mLのリン酸緩衝溶液(PBS、関東化学株式会社 73111)で洗浄後、それぞれからRNeasy Mini Kit(Qiagen 74106)を用いてRNAを抽出し、PrimeScript 1st strand cDNA Syntesis Kit(タカラバイオ 6110A)を用いてcDNAを合成し、LightCycler480(Roche)を用いてリアルタイムPCR法にてGAPDH(Applied Biosystems Hs02786624_g1)およびSYCP3(Applied Biosystems Hs00538146_m1)の遺伝子発現量を解析した。各手順はそれぞれの試薬に付属する取扱説明書に従って行った。結果を図1および図2に示す。試験した全ての細胞株において、5-AZA処理によるSYCP3の発現上昇が確認された。
BALB/cヌードマウス(10~14週齢、日本チャールズリバー)に大腸癌細胞株であるHT-29(5x105個)またはWiDr(5x105個)をマイジェクター(TERUMO SS_05M2913)にて皮内接種し、接種後5、10、15、および20日目に5-AZA(1.6μg/マウス体重g)を含むPBS溶液をマイジェクターにて200μL腹腔内投与した。コントロールには200μLのPBSを腹腔内投与した。25日目に500μg/mLのペントバルビタール(ナカライテスク 02095-04)を200μL腹腔内投与することによってマウスを安楽死させ、腫瘍組織を解剖用ハサミ(野中理化器製作所 11301)にて切除しバイオマッシャーII((株)ニッピ 320103)を用いて腫瘍組織を破砕後、試験2と同様にリアルタイムPCR法にてSYCP3およびGAPDHの遺伝子発現量を解析した。結果を図6に示す。PBSのみを投与したマウスに比べて、5-AZAを投与したマウスから回収した腫瘍組織においてSYCP3遺伝子の発現上昇が確認された。
試験1で選別されたタンパク質においてHLAに対する結合能の高いアミノ酸配列を同定するために、コンピューターアルゴリズムを用いて、日本人および欧米人において発現頻度の高いHLA-DRB1*01:01(約10%)、HLA-DRB1*04:05(約25%)、HLA-DRB1*09:01(約26%)、HLA-DRB1*15:01(約15%)、HLA-DR53(60%以上)の5種類に対する結合可能性について解析し、SYCP3-A(配列番号8)、SPESP1-B(配列番号9)およびDAZL-1C(配列番号20)を含むアミノ酸配列を選別した。
HLA-A*02:01/DRB1*01:01遺伝子組み換えマウス(A2.DR1-Tgマウス、10~16週齢;フランスパスツール研究所)にSYCP3-Aペプチド(Genscript社にて合成)を0日目と10日目に100μgをPBS100μLに溶解させてマイジェクターを用いて皮内投与した。15日目に500μg/mLのペントバルビタールを200μL腹腔内投与することによってA2.DR1-Tgマウスを安楽死させた後、開腹して所属リンパ節(dLN)および脾臓を採取し、それぞれからリンパ球および脾臓細胞を分離した。
2匹のBALB/cAnNCrlCrljマウス(Balb/c、10から16週齢;Charles river)に、リン酸緩衝溶液(PBS 関東化学株式会社 73111)に溶解したDAZL-1Cペプチド(DVQKIVESQINFHGKKLKLGPAIRKQNLC(配列番号20):Genscript社にて合成)を、0日目と10日目にマイジェクター(TERUMO SS_05M2913)を用いて100μg/50μLで皮内投与した。12日目に500μg/mLのペントバルビタール(ナカライテスク 02095-04)を200μL腹腔内投与することによってマウスを安楽死させた後、開腹して所属リンパ節(dLN)および脾臓を採取し、それぞれからリンパ球および脾臓細胞を分離した。
健常人の末梢血からリンホプレップ(Alere Technologies AS 1114547)を用いた密度勾配分離法によってPBMCを回収した。PBMCから磁気細胞分離システム(Miltenyi 130-050-201)を用いてCD14陽性細胞を分離し、50ng/mLのGM-CSF(peprotech AF-300-03)および50ng/mLのIL-4(peprotech AF-200-04)存在下で3mLのヒト細胞用培地を用いて6穴培養プレート(Falcon 353046)にて7日間培養することによって樹状細胞(Dendritic Cells;DCs)に分化させた。ヒト細胞用培地には、AIM-V培地(ThermoFisher SCIENTIFIC 0870112DK)に56℃、30分非働化したヒトAB型血清(Innovative RESEARCH IPLA-SERAB)を3%添加したものを用いた。また、PBMCから同様にしてCD4陽性T細胞(Miltenyi 130-045-101)を分離し、1x105個のCD4陽性T細胞をSYCP3-AペプチドまたはSPESP1-Bペプチド(3μg/mL)存在下で5x104個のDCsと96穴平底培養プレート(Falcon 353072)にて200μLにて共培養を始めた。7日後、CD4陽性T細胞をペプチド刺激するために、100μLの培養上清を取り除いた後、SYCP3-AペプチドまたはSPESP1-Bペプチド(3μg/mL)とガンマ線照射(40Gy)した不活化PBMC(2x105個)を100μLにて培養プレートに追加した。その2日後に50μLの培養上清を取り除き、50μLのIL-2(塩野義製薬 イムネース注35)を最終濃度が10U/mLになるように添加した。以後、活性化したCD4陽性T細胞を持続的に増殖させるために、1週間おきにSYCP3-AペプチドまたはSPESP1-Bペプチドと不活化PBMC(1x106個)を用いてCD4陽性T細胞(1x106個)を刺激し、以降に述べる各種実験を行った。
増殖してきたCD4陽性T細胞のSYCP3-Aペプチドに対する特異的反応性を調べるために、SYCP3-Aペプチド(3μg/mL)存在下で5x104個のCD4陽性T細胞と1x105個のPBMCを200μLのヒト細胞用培地を用いて96穴平底培養プレートにて共培養した。また、CD4陽性T細胞のHLA拘束性を調べるために、一部の培養系には抗DR抗体(BioLegend 307612)を、もしくは対照群として抗HLA-class I抗体(BioLegend 311412)を終濃度5μg/mLになるように添加した。24~48時間後の培養上清を100μL回収し、それに含まれるIFN-γ濃度をELISAキット(BD Biosciences 555142)を用いて付属の取扱説明書に従って測定した。
試験8と同様に、但し、SYCP3-Aペプチドの代わりにSPESP1-Bペプチドを用いて、培養上清のIFN-γ濃度を測定した。結果を図13に示す。試験した1名の健常人から2種類のSPESP1-B特異的Th細胞クローン(HK15、HK18)が樹立された。これらのクローンにおいて、抗HLA-DR抗体(aDR)を用いた場合にペプチド特異的IFN-γ産生が抑制されたことから、SPESP1-BペプチドはHLA-DR拘束的にTh細胞を刺激していることが確認された。また、試験8と同様にL-DR53を用いた実験により、SPESP1-BはHLA-DR53に結合し、Th細胞を刺激していることが確認された。
試験7と同様に、但し、SYCP3-AペプチドまたはSPESP1-Bペプチドの代わりにDAZL-1断片ペプチドp11、p12またはp13(表4)を用いて、DAZL-1ペプチド特異的Th細胞を誘導した。
SYCP3-Aペプチド特異的CD4陽性T細胞の癌細胞株に対する反応性を調べるために、DR53陽性(WiDr、Lu65、Calu1)癌細胞株を選別した。各癌細胞株を、10μMの5-AZAとともに、癌細胞株の細胞表面上にHLAクラスIIを発現させるための500U/mLのIFN-γ(塩野義製薬 イムノマックス-γ注50)を含む2mLの完全培地を用いて、6穴培養プレートにて3日間培養した。各癌細胞株の培養プレートをPBSでよく洗浄し、5mMのEDTA(エチレンジアミン四酢酸 ナカライテスク 14347-21)を1mL用いて各細胞株を遊離させ、各癌細胞株を回収した。試験8と同様に、1x104個の各細胞株と5x104個のCD4陽性T細胞を200μLのヒト細胞用培地を用いて96穴平底培養プレートにて共培養した。また、HLA-DR依存的な反応性であることを調べるために培養系に抗HLA-DR抗体(終濃度5μg/mL)を添加した。24時間後の培養上清を100μL回収し、それに含まれるIFN-γ濃度をELISAキットを用いて測定した。
BALB/cヌードマウス(10~14週齢、日本チャールズリバー)に肺癌細胞株であるLu65(3x106個)をマイジェクターにて皮内接種し、接種後7、12、17、および22日目に5-AZA(150nmol/マウス体重g)を含むPBS溶液をマイジェクターにて200μL腹腔内投与した。コントロールには200μLのPBSを腹腔内投与した。一方、接種後13、20、27日目にSYCP3特異的ヒトTh細胞(3~5x106個)を200μLで尾静脈投与し、コントロールには200μLのPBSを尾静脈投与した。腫瘍の表面積を継時的に測定した。結果を図16に示す。5-AZAの単剤投与では効果は認められなかったが、SYCP3特異的ヒトTh細胞との併用投与によって腫瘍増殖抑制効果が認められた。
