Abstract
Description
(1)GDF6タンパク質またはmiR-17ファミリーメンバーを含む、老化に関連する状態を処置するための組成物。
(2)GDF6タンパク質またはmiR-17ファミリーメンバーのヒト発現用ベクターを含む、老化に関連する状態を処置するための組成物。
(3)GDF6タンパク質を分泌する細胞を含む、老化に関連する状態を処置するための組成物。
(4)GDF6タンパク質またはmiR-17ファミリーメンバーを過剰発現した細胞。
(5)間葉系幹/間質細胞または血球細胞にGDF6タンパク質またはmiR-17ファミリーメンバーを過剰発現させて得られる、上記(4)に記載の細胞。
(6)間葉系幹/間質細胞または血球細胞が老化した細胞である、上記(5)に記載の細胞。
(7)ヒト細胞である、上記(4)~(6)のいずれかに記載の細胞。
(8)細胞が、上記(4)~(7)のいずれかに記載の細胞である、上記(3)に記載の組成物。
(9)GDF6タンパク質が、
(i)配列番号2または3に示されるアミノ酸配列を有するか、
(ii)配列番号2または3に示されるアミノ酸配列と90%以上の相同性を示す配列を有するか、
(iii)配列番号2または3に示されるアミノ酸配列において、1個~10個のアミノ酸の欠失、挿入および/または置換を有するアミノ酸配列を有するか、または
(iv)配列番号2または3に示されるアミノ酸配列をコードするDNA配列に対して相補的な配列を有する核酸と、ストリンジェントな条件でハイブリダイズ可能な配列によりコードされるアミノ酸配列を有する、
上記(1)~(3)および(8)のいずれかに記載の組成物。
(10)miR-17ファミリーメンバーが、
(i)配列番号4に示される核酸配列を有するか、
(ii)配列番号4に示される核酸配列と90%以上の同一性を有するか、または、
(iii)配列番号4に示される核酸配列において、1個~10個の塩基の欠失、挿入および/または置換を有する核酸配列を有する、
上記(1)、(2)または(8)に記載の組成物。
(11)老化に関連する状態が、筋肉の老化、造血系の老化、神経系の老化、血管系の老化、皮膚組織の老化、毛髪の老化または細胞老化である、上記(1)~(3)および(8)~(10)のいずれかに記載の組成物。
(12)老化に関連する状態が、老化に伴う以下の状態:筋肉の損傷の回復力の減少、リンパ球の減少、神経幹/前駆細胞の減少、血管壁の硬化、血管内腔の縮小、血管網の減退、皮膚組織の弾力の低下、皮膚組織の菲薄化、毛つやの低下、未分化細胞の減少またはSASP因子の分泌上昇である、上記(1)~(3)および(8)~(10)のいずれかに記載の組成物。
(13)GDF6タンパク質が、
(i)配列番号2または3に示されるアミノ酸配列を有するか、
(ii)配列番号2または3に示されるアミノ酸配列と90%以上の相同性を示す配列を有するか、
(iii)配列番号2または3に示されるアミノ酸配列において、1個~10個のアミノ酸の欠失、挿入および/または置換を有するアミノ酸配列を有するか、または
(iv)配列番号2または3に示されるアミノ酸配列をコードするDNA配列に対して相補的な配列を有する核酸と、ストリンジェントな条件でハイブリダイズ可能な配列によりコードされるアミノ酸配列を有する、
上記(4)~(7)のいずれかに記載の細胞。
(14)miR-17ファミリーメンバーが、
(i)配列番号4に示される核酸配列を有するか、
(ii)配列番号4に示される核酸配列と90%以上の同一性を有するか、または、
(iii)配列番号4に示される核酸配列において、1個~10個の塩基の欠失、挿入および/または置換を有する核酸配列を有する、
上記(4)~(7)のいずれかに記載の細胞。
(i)配列番号3で示されるアミノ酸配列を有するタンパク質であり得る。
あるいは、他の好ましい態様では、本発明で用いられるGDF6タンパク質は、
(ii)配列番号3に示されるアミノ酸配列と90%以上、より好ましくは95%以上、さらに好ましくは98%以上、さらにより好ましくは99%の相同性(または同一性)を示す配列を有するか、または、
(iii)配列番号3に示されるアミノ酸配列において、1個~10個、より好ましくは1個~5個、さらに好ましくは1個~3個、さらにより好ましくは1個のアミノ酸の欠失、挿入および/または置換を有するアミノ酸配列を有する、タンパク質であり得る。本発明では、GDF6タンパク質の活性化型フォームはホモ二量体を形成し得る。
(i)配列番号2で示されるアミノ酸配列を有するタンパク質であり得る。
あるいは、他の好ましい態様では、本発明で用いられるGDF6タンパク質は、
(ii)配列番号2に示されるアミノ酸配列と90%以上、より好ましくは95%以上、さらに好ましくは98%以上、さらにより好ましくは99%の相同性(または同一性)を示す配列を有するか、または、
(iii)配列番号2に示されるアミノ酸配列において、1個~10個、より好ましくは1個~5個、さらに好ましくは1個~3個、さらにより好ましくは1個のアミノ酸の欠失、挿入および/または置換を有するアミノ酸配列を有する、タンパク質であり得る。本発明では、GDF6タンパク質の前駆体フォームはホモ二量体を形成し得る。前駆体フォームが活性化型フォームに変換されるためには、配列番号2に示されるアミノ酸配列の331番~335番に対応するアミノ酸の配列が保存されていることが好ましい。従って、本発明で用いられるGDF6タンパク質が前駆体フォームとして提供される場合には、配列番号2に示されるアミノ酸配列の331番~335番に対応するアミノ酸の配列が保存されているか、または、RXXRモチーフ(ここでXは任意のアミノ酸を意味する)が保存されているGDFタンパク質を用いることが好ましい。
(i)配列番号4~21のいずれかの核酸配列を有するか、
(ii)配列番号4~21のいずれかに示される核酸配列と90%以上の同一性を有するか、または、
(iii)配列番号4~21のいずれかに示される核酸配列において、1個~10個、好ましくは1~5個、より好ましくは1~3個、さらに好ましくは1~2個、さらにより好ましくは1個の塩基の欠失、挿入および/または置換を有する核酸配列を有する、RNAとすることができる。例えば、miR-17としては、
(i)配列番号4に示される核酸配列を有するか、
(ii)配列番号4に示される核酸配列と90%以上の同一性を有するか、または、
(iii)配列番号4に示される核酸配列において、1個~10個、好ましくは1~5個、より好ましくは1~3個、さらに好ましくは1~2個、さらにより好ましくは1個の塩基の欠失、挿入および/または置換を有する核酸配列を有する、RNAとすることができる。miR-17ファミリーメンバーがGDF6タンパク質を発現誘導するか否かは、例えば、miR-17ファミリーメンバーを過剰発現する細胞またはその培養上清をウェスタンブロット法により分析することにより評価できる。
また、miR-17ファミリーメンバーを発現するベクターは当業者に周知の方法で作製できる。マイクロRNAは、当業者に周知の方法により調製することができる。例えば、抗Ago2抗体を用いて細胞抽出物からマイクロRNAを精製する方法が知られている。
本実施例では、MSCの老化に伴う遺伝子発現の変動を解析した。
本実施例では、実施例1で示された老化に伴うMSCの能力の低下に関連する因子を同定した。
実施例2では、miR-17を強制発現させたOld MSCにおいて、老化現象に特徴的であった骨形成能や脂肪形成能が部分的に回復した。本実施例では、このように老化に伴う能力低下を回復させたMSCを移植してこれがレシピエントにおけるリンパ球再生能に及ぼす影響を確認した。
実施例3の結果から、MSCは老化を回復する分泌性因子を放出して老齢マウスに対して加齢による生理機能の低下の回復および/または改善作用を発揮させることが明らかとなった。本実施例では、加齢による生理機能の低下の回復および/または改善作用を担う分泌性因子を同定した。
(1)発現量が、Young MSCと比較してOld MSCにおいて0.7倍未満である
(2)発現量が、Young MSCと比較して陰性対照(miR-LacZ)を導入したOld MSCにおいて0.7倍未満である
(3)発現量が、Young MSCと比較してmiR-17を強制発現させたOld MSCにおいて0.7倍以上である
(4)発現量が、陰性対照を導入したOld MSCと比較してmiR-17を強制発現させたOld MSCにおいて1.5倍を超える
本実施例では、MSCを用いてインビトロでGdf6をノックダウンすることにより、Gdf6の生理的意義を分析した。本実施例ではまた、インビボでGdf6を老齢マウスに発現させて老化現象に対する効果を検証した。
本実施例では、細胞老化に伴うSASP因子の分泌をGDF6が抑制することができるかどうかを調べた。
実施例5および6では、老齢マウス自体にレンチウイルスを投与してGDF6をマウスに強制発現させたが、本実施例では、GDF6を強制発現させた末梢血を老齢マウスに投与して老化に対する効果を検証した。
以上より、老齢マウスにhGDF6を過剰発現する細胞を投与することで、hGDF6の遺伝子発現用ベクターの導入と同様の効果を得ることが確認された。
次に、前述の実施例と同じhGDF6をコードするDNAを発現可能に連結したレンチウイルスを若齢マウスの腹腔内に1日1回の頻度として3日連続で投与し、老化に伴うマウスの毛の減少および毛のつやについて経過観察した。陰性対象としては、ウイルス投与をしていないマウスおよびGDF6に代えてEGFPを発現させたマウスを用いて、同ケージ内で飼育した。結果は図12に示される通りであった。図12に示されるように、投与から2カ月後及び8カ月後のそれぞれにおいて、hGDF6を発現させたマウスでは、陰性対照と比較して背中全体で毛および毛のつやが維持されていることが示された。
Claims (14)
- GDF6タンパク質またはmiR-17ファミリーメンバーを含む、老化に関連する状態を処置するための組成物。
- GDF6タンパク質またはmiR-17ファミリーメンバーのヒト発現用ベクターを含む、老化に関連する状態を処置するための組成物。
- GDF6タンパク質を分泌する細胞を含む、老化に関連する状態を処置するための組成物。
- GDF6タンパク質またはmiR-17ファミリーメンバーを過剰発現した細胞。
- 間葉系幹/間質細胞または血球細胞にGDF6タンパク質またはmiR-17ファミリーメンバーを過剰発現させて得られる、請求項4に記載の細胞。
- 間葉系幹/間質細胞または血球細胞が老化した細胞である、請求項5に記載の細胞。
- ヒト細胞である、請求項4~6のいずれか一項に記載の細胞。
- 細胞が、請求項4~7のいずれか一項に記載の細胞である、請求項3に記載の組成物。
- GDF6タンパク質が、
(i)配列番号2または3に示されるアミノ酸配列を有するか、
(ii)配列番号2または3に示されるアミノ酸配列と90%以上の相同性を示す配列を有するか、
(iii)配列番号2または3に示されるアミノ酸配列において、1個~10個のアミノ酸の欠失、挿入および/または置換を有するアミノ酸配列を有するか、または、
(iv)配列番号2または3に示されるアミノ酸配列をコードするDNA配列に対して相補的な配列を有する核酸と、ストリンジェントな条件でハイブリダイズ可能な配列によりコードされるアミノ酸配列を有する、
請求項1~3および8のいずれか一項に記載の組成物。 - miR-17ファミリーメンバーが、
(i)配列番号4に示される核酸配列を有するか、
(ii)配列番号4に示される核酸配列と90%以上の同一性を有するか、または、
(iii)配列番号4に示される核酸配列において、1個~10個の塩基の欠失、挿入および/または置換を有する核酸配列を有する、
請求項1、2または8に記載の組成物。 - 老化に関連する状態が、筋肉の老化、造血系の老化、神経系の老化、血管系の老化、皮膚組織の老化、毛髪の老化または細胞老化である、請求項1~3および8~10のいずれか一項に記載の組成物。
