- Microtubules perform numerous cellular functions, such as cell migration, cell division, cytoskeletal maintenance and intracellular transport.
- the main protein component of microtubules is tubulin.
- the rapid growth of cancer cells is highly dependent on tubulin polymerization / depolymerization, and therefore tubulin is an excellent target for anticancer drug development.
- Intervention into the microtubule conjugates, by inhibiting tubulin polymerization or blocking microtubule degradation increases the number of cells trapped in the metaphase, resulting in cell death. Inhibiting the function of microtubules with drugs targeting tubulin is a recognized approach to anticancer therapy.
- Microtubule stabilizers include taxanes, paclitaxel, docetaxel, and the like, which inhibit depolymerization of microtubules and enhance microtubule polymerization. Most microtubule stabilizers bind to the taxane binding site or the overlapping site of ⁇ -tubulin.
- the second group of microtubule destabilizers include colchicine, vinca alkaloid, and the like, which inhibit microtubule polymerization and mostly bind to colchicine binding sites or vinca binding sites.
- two classes of drugs targeting microtubules act at lower concentrations than drugs that affect microtubule polymers.

- the indole derivative of the present invention WCI-1031 cells were collected in the divider at a concentration of 0.5 to 1.0 ⁇ M, it was confirmed that the number of cells collected in the divider is more than 50%. Therefore, it was confirmed that the indole derivative of the present invention inhibits the cell cycle in the G2, M phase.
- HeLa cells were treated with DMSO or WCI-1031 (0.1 ⁇ M, 0.5 ⁇ M, 1.0 ⁇ M) for 16 hours. After fixing the cells, stained with anti-tubulin antibody and Alexa Fluor 488, stained with anti-centrosome antibody, Texas Red, and immuno-stained nuclei using Hoechst 33342 to confirm ⁇ -tubulin, centrosome and DNA ( 1).
- the indole derivative of the present invention is an agent for depolymerizing the microtubules.
- the cell-derived skin cancer cell line A431 which was frozen in liquid nitrogen, was thawed and then cultured. The culture of the cells was incubated for a suitable period of time in a CO 2 incubator (Forma, USA) at a temperature of 37 °C and 5% CO 2 concentration.
- WCI-1031 (Example 1), an indole derivative of the present invention, was used as a test substance, and 5-FU and colchicine were used as positive controls.
- the prepared materials were repeatedly intraperitoneally and orally administered 0.2 ml (10 ml / kg) per 20 g of the mouse to the schedule below.
- the average tumor size of each group from 57.0 mm 3 to 23rd day was measured in three directions using a vernier caliper, and then expressed as a formula of length ⁇ width ⁇ height / 2.
- the WCI-1031 (10, 20 mg / kg) intraperitoneal group showed 9.3% and 37.2% (p ⁇ 0.001) of tumor growth inhibition compared to the solvent control group, respectively.
- tumor growth inhibition was 20.7% (p ⁇ 0.01) and 17.6% (p ⁇ 0.05), respectively (FIGS. 3 and 4).
- A431 tumors were excised and weighed 16 days after the initiation of drug administration. As compared with the solvent control group, WCI-1031 (10, 20 mg / kg) group received 10.7% and 38.0% (p ⁇ 0.001) tumors, respectively. Weight loss appeared. In the positive control 5-FU and colchicine groups, tumor weight was reduced by 20.8% (p ⁇ 0.05) and 17.6% (p ⁇ 0.05), respectively (FIGS. 5 and 6).
- a solid sample ( ⁇ 1 mg) of the test substance was placed in a Whatman Syringeless filter (PVDF membrane, 0.45 um pore size) and mixed with 0.5 ml of phosphate buffer (pH 6.5 or 7.4). The mixture was sonicated for 15 minutes and vortex mixed for 1 hour at room temperature. The equilibrated mixture was filtered and the filtrate was analyzed with a multi-wavelength UV plate reader (FIGS. 8 and 9).
- SIWV small intestinal water volume ( ⁇ 250 ml)
- SITT small intestinal transit time ( ⁇ 270 min)
