Abstract

A method for carrying out a nucleic acid amplification reaction to amplify a specific sequence which includes a polymorphism, which method comprising carrying out a nucleic acid amplification reaction using a nucleic acid amplification reaction mixture which comprises a nucleic acid primer having a 3'end that binds at or near the site of the polymorphism, and a set of nucleotides able to effect amplification of the target sequence and wherein the composition of the nucleotides is selected so as to enhance the specificity of the primer for the specific sequence. In particular, a substitute nucleotide or modified nucleotide analogue that competes with the natural nucleotide for incorporation is included in the reaction at a relatively high concentration, and/or the concentration of one or more of the nucleotides is reduced.
