- the mobile phase from which the target component has been removed is discarded through the flow path 12 and the flow path 13 to the waste liquid port.
- the control unit 30 When the solution is supplied to the trap column 8 only for a predetermined time or until the solution prepared in the solution container 1 runs out, the control unit 30 next selects the pure water container 2 (c port), the flow path 5 (a port), The switching valve 4 is switched so as to be connected. Then, the liquid feed pump 6 sucks the pure water in the pure water container 2 and introduces it into the trap column 8. Thereby, undesired water-soluble substances such as salts adhering to the adsorbent during the collection of the target component are removed from the trap column 8. By supplying pure water, the mobile phase accumulated in the trap column 8 immediately before the start of the supply is replaced with water, and the trap column 8 is filled with water. Since the target component collected in the adsorbent hardly elutes in water due to the strong adsorption action, the state of being collected in the trap column 8 is maintained at this point.
- control unit 30 moves the sorting head 16 above the predetermined collection container 21 specified in advance by the triaxial drive mechanism 29, and the outlet ends of the solution connecting pipe 17A and the gas connecting pipe 17B
- the sorting head 16 is lowered to a predetermined height so as to be fitted to the inlet ends of the introduction pipe 20A and the gas introduction pipe 20B, respectively (FIG. 3 (b)).
- the switching valve 4 is switched so as to connect the elution solvent container 3 (d port) and the flow path 5 (a port).
- the liquid feed pump 6 sucks the dichloromethane in the elution solvent container 3 and starts to introduce it into the trap column 8.
- the liquid feeding flow rate of the liquid feeding pump 6 is set to a predetermined flow rate smaller than that of the previous solution or pure water.
- the control unit 30 instructs the temperature adjustment unit 28 to specify the target temperature and starts heating the temperature adjustment block 27, thereby starting to heat the collection container 21.
- the target temperature may be, for example, about the same as or slightly higher than the boiling point of dichloromethane, and may be about 40 to 45 ° C.
- dichloromethane When dichloromethane is introduced into the trap column 8, the interface between dichloromethane and water gradually rises with almost no mixing with the water present in the trap column 8. That is, dichloromethane gradually accumulates from the bottom of the trap column 8 while pushing up water. On the other hand, the pushed-up water overflows from the outlet end at the upper end of the trap column 8, passes through the switching valve 15 and reaches the waste liquid port from the flow path 13. On the other hand, since dichloromethane has a strong dissolution power, the target component collected in the trap column 8 is dissolved in the dichloromethane accumulated in the trap column 8.
- the switching valve 15 is switched from the flow path 13 (b port) to the flow path 14 (c port), and separation of the target component is started. . Further, the control unit 30 causes the gas supply unit 23 to start supplying nitrogen gas (or other inert gas). The gas delivered from the gas supply unit 23 is introduced into the gas introduction pipe 20B via the flow path 22 and the gas connection pipe 17B, and begins to blow out from the tip of the gas introduction pipe 20B.
- the solution sent from the trap column 8, that is, dichloromethane containing the target component is dropped from the tip of the solution introduction pipe 20A through the flow path 12, the flow path 14, and the solution connection pipe 17A. At the time of this dropping, the solution is sheared by the gas flow blown from the surroundings, and is scattered as fine droplets.
- the recovery container 21 is heated to the same level as the boiling point of dichloromethane by heat conduction from the temperature control block 27 using the heater 25 as a heat source. Therefore, when a fine droplet of the solution adheres to the inner peripheral wall surface or inner bottom wall surface of the collection container 21, the solvent (dichloromethane) in the droplet immediately evaporates and the target component remains as a powder. Thus, the powdery target component is deposited on the inner peripheral wall surface and inner bottom wall surface of the collection container 21.
- the sorting head 16 is raised and the connection state with the cap 20 is released.
- the sorting head 16 is moved to a position where the next collection container 21 is located, and the same processing is performed.
- dichloromethane sent to the flow path 11 is introduced into the flow path 22 and is introduced into the gas introduction pipe 20B through the gas connection pipe 17B.
- dichloromethane since dichloromethane has a strong dissolution power, the precipitate that has entered the gas introduction pipe 20B is dissolved and washed away by the dichloromethane introduced into the gas introduction pipe 20B.
- the switching valve 7 is switched from the flow path 11 (c port) to the flow path 10 (b port), the supply of gas from the gas supply unit 23 is resumed, and the above-described gas spraying evaporation / drying process is continued. To do.
- the solute (precipitate) deposited at the tip of the solution introduction tube grows along the outer periphery of the solution introduction tube 20A and can enter the gas introduction tube 20B by the following method.
- the inventor of the present invention has a limit in the distance that the precipitate grows along the outer periphery of the solution introduction tube 20A, and does not grow beyond a certain distance.
- the distance w of this limit depends on the surface tension of the solvent and the solvent. It has been found through experiments that the angle substantially depends on the contact angle with the solution introduction tube 20A.
- PTFE polytetrafluoroethylene
- dichloromethane was used as a solvent.
- the contact angle to the solution introduction tube 20A is 114 °.
- the inner diameter of the solution introduction tube 20A was 0.8 mm
- the flow rate of the solution flowing through the solution introduction tube 20A was 0.1 mL / min
- the flow rate of the gas flowing through the gas introduction tube 20B was 1.6 L / min.
- the growth limit distance w greatly depends on the contact angle of the solvent and the surface tension (correlated with the contact angle.
- the tip of the solution introduction tube 20A and the gas introduction tube If the distance d between the tips of 20B is set to 9 mm or more, it is possible to prevent the outlet of the gas introduction pipe from being blocked by the precipitated and grown solute. 14 mm, which is about 1.5 times the growth limit distance w confirmed in the experiment (FIG. 4A).
- the contact angle is 96 °.
- the precipitates propagate up to the outer periphery of the solution introduction tube 20A up to 12 mm. I grew up.
- the precipitate grows downward as well.
- the precipitate grown downward is peeled off from the tip of the solution introduction tube 20A by its own weight when grown to a certain extent.
- the precipitate is separated.
- the bottom of the solution introduction tube 20A and the bottom of the collection container 21 are connected in a columnar shape.
- the amount of precipitates increased as the cross-sectional area of the outlet end face of the solution introduction tube 20A increased.
- FIG. 4C when the outlet end face has a shape that is cut obliquely, the cross-sectional area of the outlet end face increases, and the amount of precipitation increases. Therefore, it is desirable that the outlet end face of the solution introduction tube 20A be cut perpendicularly to the longitudinal direction of the solution introduction tube 20A so that the cross-sectional area is minimized (FIG. 4B).
- the solution introduction tube 20A should have a high contact angle at least on the outer peripheral surface thereof.
- a fluorine-based resin material can be used as a constituent material of the solution introduction tube 20A.

It can be provided as a fixing means for fixing to the block 27.
- a set of the screw thread 33A and the screw groove 33B is referred to as a first screw 33
- a set of the screw thread 34A and the screw groove 34B is referred to as a second screw 34.
- the cap 35 When the cap 35 is fitted into the collection container 21 and the cap 35 is rotated, the cap 35 is fixed to the collection container 21 by the first screw 33 and attached to the upper opening of the collection container 21. This is a structure often used in the past.
- the collection container 21 with the cap 20 attached thereto is inserted into the temperature control block 27 by the first screw 33 and the cap 20 is rotated together with the collection container 21, the cap 20 and the temperature control block 27 is fixed by the second screw 34. Since the temperature control block 27 is a part of the container rack 24, the weight of the container rack 24 as a whole prevents the collection container 21 from being lifted together with the cap 20 when the sorting head 16 is raised.
- FIG. 5A shows a state where none of the collection container 21, the cap 20, and the temperature control block 27 is mounted or inserted.
- the recovery container 21 is accommodated in the temperature control block 27.
- the collection container 21 is not fixed to the temperature control block 27.

- This preparative purification apparatus is different from the preparative purification apparatus of the first embodiment shown in FIG. 1 in that the cap 35 does not include the solution introduction pipe 20A, the gas introduction pipe 20B, the exhaust port 20C, and the filter 20D.
- a needle 37 having a double pipe structure of a solution introduction pipe and a gas introduction pipe is used.
- the cleaning liquid is fed through the flow path 22 for supplying gas in the same procedure as in the first embodiment. . Thereby, even if the precipitate enters the inside of the gas introduction pipe, it can be washed away.
- the needle 37 pierces the two septa 52A of the cap 35 shown in FIG. .
- the sorting head 16 is raised together with the needle 37, and the needle 37 is pulled out from the cap 35.

Abstract
