Abstract

L'invention concerne des procédés, des moyens et des coffrets pour la détection et l'analyse quantitative d'analytes chimiques et biologique à l'aide d'une amplification de signal catalytique. L'invention concerne des procédés, des coffrets et des réactifs pour détecter une molécule cible ou faire intéragir des molécules cibles dans un échantillon sur la base d'une amplification de signal catalytique, comprenant les étapes consistant à (i) se procurer au moins une première sonde non fluorescente et une seconde sonde non fluorescente aptes à se lier à des sites de liaison distincts sur la (les) molécule(s) cible(s), la première sonde étant dotée d'un premier ligand et la seconde sonde étant dotée d'un second ligand, les premier et second ligands pouvant former un complexe catalytique en présence d'un composé catalytique et le complexe catalytique pouvant catalyser une réaction chromogène; (ii) mettre en contact un échantillon que l'on sait ou que l'on suspecte de contenir la (les molécule(s) cible(s) avec lesdites au moins première et seconde sondes non fluorescentes dans des conditions qui permettent la liaison des sondes à (aux) molécule(s) cible(s) de telle sorte que, lors de la liaison des sondes à (aux) molécule(s) cible(s), les premier et second ligands sont amenés en proximité proche l'un de l'autre et conjointement avec le composé catalytique forment un complexe catalytique immobilisé sur la (les) molécule(s) cible(s); et (iii) détecter la formation du complexe catalytique sur la base d'une conversion non enzymatique d'un colorant précurseur soluble dans l'eau en un colorant rapporteur chromophore.
