Abstract
La presente invención proporciona un método in vitro de preparación de un tejido artificial, el tejido artificial obtenible por dicho método, la composición farmacéutica que lo comprende y el uso de este tejido o de esta composición para incrementar, restaurar o sustituir parcial o totalmente la actividad funcional de un tejido o un órgano dañado, preferiblemente piel, vejiga, uretra, córnea, mucosa oral, conjuntiva, pared abdominal, tímpano, faringe, laringe, intestino, peritoneo, ligamento, tendón, hueso, meninge o vagina. El método de la invención comprende los pasos de añadir fibrinógeno a células aisladas, preferiblemente, a fibroblastos o queratocitos, añadir un agente antifibrinolítico a la mezcla, añadir además un factor de coagulación, una fuente de calcio y trombina, añadir un polisacárido, preferiblemente agarosa, cultivar células, preferiblemente células epiteliales o células madre del cordón umbilical, en o sobre el producto resultante e inducir la nanoestructuracion de dicho producto. El método contempla también un paso intermedio que consiste en la adición de fibronectina tras el agente fibrinolítico, y otro paso en el que se añade colágeno después de la agarosa.
