- the proteins are separated by SDS-PAGE electrophoresis. After semi-dry transfer onto a PVDF membrane (Immobilon-P Millipore) according to a standard protocol, the proteins are incubated with the anti-DJ-1 primary antibody (Abeam, ab 4150) overnight at 4°C. The second incubation is then performed with the secondary antibody (goat anti-mouse IgG-HRP conjugate; Bio-Rad) directed against the primary antibody, for 1 hour 30 minutes at room temperature. The presence of the protein DJ-I on the membrane is revealed by immunodetection using the ECL plus kit (Amersham). The membrane is then stained with Amidoblack to detect the total proteins present on the membrane. The image is acquired using a Fluor S Max machine (Bio-Rad) and the bands are quantified using the Quantity-one software (Bio-Rad).
- Results The results are expressed in delta cnt*mm 2 of the protein of interest/delta cnt*mm 2 of the total proteins.

Abstract
La présente invention porte sur l'utilisation, notamment l'utilisation cosmétique et/ou thérapeutique, de la protéine DJ-1, de polypeptides issus de cette protéine ou d'analogues correspondants, d'une séquence d'acide nucléique codant pour un tel polypeptide ou d'un modulateur de l'activité, de la stabilité ou de l'expression d'un tel polypeptide, notamment pour la prévention et/ou le traitement des signes d'une sécheresse cutanée. L'invention porte également sur l'utilisation de la protéine DJ-1, de polypeptides issus de cette protéine ou d'analogues correspondants, ou d'une séquence d'acide nucléique codant pour un tel polypeptide, en tant que marqueur pour évaluer l'état de sécheresse d'un épithélium.
