- Example 2 -Inhibition of a kidney tumor cell line Adherent cells are plated approximately 16-24 hours before the day of the experiment in 180 ⁇ l growth media. On the day of the experiment, the plated adherent cells are analyzed and counted and suspension cells are plated in 180 ⁇ l growth media. The compounds and vehicle are prepared. The resulting solutions are added to the cells. The cells are incubated at 37 0 C, 5% CO 2 for 48 hours. The media is then aspirated. The plate containing the cell suspension is then spun at 1500 RPM for 10 minutes. The media is slowly removed using a multichannel pipettor. 200 ⁇ l of MTT is added to each well to a concentration of 0.863mg/ml MTT in the growth media.
