Abstract
Description
このようなグルタチオンの品質低下を抑制する方法として、グルタチオンの粉末の粒子表面を被覆する方法(特許文献1および2参照)、シクロデキストリンを添加する方法(特許文献1および3参照)等が知られているが、操作が煩雑である、効果が弱い等の問題がある。
このようにグルタチオンとアルギニンはいずれも多様な生理作用を有しており、免疫賦活作用等の共通する生理作用もあることから、グルタチオンとアルギニンとを同時に摂取することにより、それぞれの生理作用について、相加効果だけでなく、相乗効果が得られることも期待できる。
(1)グルタチオンおよびアルギニンと有機酸とを共存させることを特徴とする、グルタチオンおよびアルギニンを含有する固体組成物におけるグルタチオンの保存安定性向上方法。
(2)固体組成物が固形製剤である、上記(1)の方法。
(3)有機酸が、クエン酸、酒石酸、アスコルビン酸およびリンゴ酸から選ばれる有機酸である、上記(1)または(2)の方法。
(4)有機酸が、クエン酸または酒石酸である、上記(1)~(3)のいずれか1つの方法。
(5)グルタチオンおよびアルギニンとクエン酸または酒石酸とを含有することを特徴とする固体組成物。
(6)グルタチオンおよびアルギニンとクエン酸または酒石酸とを含有することを特徴とする固形製剤。
本発明に用いられるグルタチオンは、還元型(L-γ-グルタミル-L-システイニルグリシン)および酸化型(グルタチオンジスルフィド)のいずれであってもよい。
本発明におけるアルギニンは、L体、D体のいずれでもよいが、L体が好ましい。 アルギニンは、塩酸塩、硫酸塩、リン酸塩等の無機酸の塩であってもよいが、フリー体が好ましく用いられる。
グルタチオンおよびアルギニンと共存させる有機酸としては、常温で固体の有機酸であればいずれの有機酸であってもよいが、医薬品、飲食品、飼料等への使用という観点から、乳酸、酒石酸、アスコルビン酸、リンゴ酸、マロン酸、コハク酸、フマル酸、マレイン酸、クエン酸等があげられ、クエン酸、酒石酸、アスコルビン酸およびリンゴ酸が好ましく、クエン酸、酒石酸およびアスコルビン酸がより好ましく、クエン酸および酒石酸がさらに好ましく用いられる。
グルタチオンおよびアルギニンと共存させる有機酸の量は、アルギニン1重量部に対して通常0.1~2重量部、好ましくは0.2~1.2重量部、さらに好ましくは0.4~0.8重量部である。
グルタチオンおよびアルギニンと有機酸とを共存させる際、グルタチオンの保存安定性に悪影響を及ぼさない、医薬品、食品または飼料の分野で通常用いられる物質をさらに共存させてもよい。
賦形剤としては、マルトース、トレハロース、マンニトール、還元麦芽糖水飴、ラクチトール、キシリトール、ソルビトール、エリスリトール、結晶セルロース、低置換度ヒドロキシプロピルセルロース等があげられる。
結合剤としては、ポリビニルピロリドン、プルラン、メチルセルロース、ヒドロキシプロピルセルロース、ポリビニルアルコール、ゼラチン、寒天等があげられる。
甘味剤としては、サッカリンナトリウム、グリチルリチン二カリウム、アスパルテーム、ステビア、ソーマチン、スクラロース、グルコース、フルクトース、サッカロース等があげられる。
着色剤としては、食用黄色5号、食用赤色2号、食用青色2号、カロチノイド色素、トマト色素等があげられる。
抗酸化剤としては、トコフェロール、塩酸システイン等があげられる。
流動化剤としては、リン酸カルシウム、リン酸水素カルシウム、微粒二酸化ケイ素等があげられる。
上記のようにこれらの物質を共存させて得られる本発明の固体組成物の水分含量は5重量%を超えないようにすることが好ましく、3重量%を超えないようにすることがさらに好ましい。
本発明の保存安定性向上方法により、グルタチオンおよびアルギニンを含有する固体組成物中のグルタチオンの品質の低下を抑制し、保存安定性を向上させることができる。
グルタチオンの保存安定性は、所定の条件下で保存する前の固体組成物中のグルタチオンの含有量に対する保存後のグルタチオンの含有量の百分率として知ることができる。
例えば、散剤は、粉末状の製剤原料を混合機を用いて混合するか、または製剤原料を粉砕機等により粉砕し、混合機等を用いて混合して製造することができる。
錠剤は、打錠機を用いて製剤原料を打錠して製造することができる。
なお、顆粒剤、錠剤は調製後、糖、糖アルコール等を用いるシュガーコーティング、高分子を用いるフィルムコーティング等を施して用いてもよい。
カプセル剤は、散剤または顆粒剤を調製後、ハードカプセルに充填して製造することができる。
本発明のグルタチオンおよびアルギニンを含有する固形製剤は、医薬品、食品または飼料分野において利用することができるが、ヒトまたは非ヒト動物に投与する際は、グルタチオンおよびアルギニンまたはその塩として、一日あたり、100mg~20gとなるように投与することが好ましい。
粉末1~4を、各々アルミパウチ袋に入れ、密封し、60℃で2週間保存した。2週間後、アルミパウチ袋から、少量をサンプリングし、グルタチオンの含有量を高速液体クロマトグラフィーにより定量した。
カラム:Nucleosil 10-C18 4.6mmΦX250mm(ジーエルサイエンス社製)
移動相:2.84gのギ酸アンモニウムを水に溶解後、ギ酸でpH4に調整し、水で全量を1800mlとした後、メタノール200mlを加えて撹拌し、脱気を行なって調製したもの

移動相流量:1.0ml/分
保存前の各粉末(粉末1~4)中のグルタチオンの含有量および保存後の各粉末(粉末1~4)中のグルタチオンの含有量から、グルタチオンの残存率を算出した。
結果を第1表に示す。
(2)0.5gのグルタチオンおよび10gのL-アルギニンに、それぞれ、クエン酸を第2表記載の量添加、混合し、攪拌して粉末5~7を得た。
粉末5~7を各々アルミパウチ袋に入れ、密封し、60℃で2週間保存した。2週間後、各粉末中のグルタチオンの残存率を上記(1)記載の方法に準じて算出した。
結果を第2表に示す。
(3)0.5gのグルタチオンおよび5gのL-アルギニンに、それぞれ、第3表記載の有機酸を第3表記載の量添加、混合し、攪拌して粉末8~11を得た。
粉末8~11を各々アルミパウチ袋に入れて、密封し、60℃で2週間保存した。2週間後、各粉末中のグルタチオンの残存率を上記(1)記載の方法に準じて算出した。
結果を第3表に示す。
