Abstract

Le procédé décrit permet de produire un polypeptide ou un peptide, ou un acide nucléique codant pour le polypeptide ou le peptide. Ledit polypeptide ou peptide comprend une séquence peptidique modifiée ayant une affinité de liaison accrue pour une molécule de la Classe I du CMH et/ou générant une réponse immunitaire en lymphocytes T CD8+ plus élevée. Le procédé comprend les étapes consistant à : identifier une séquence peptidique (« peptide parent ») d'un épitope de lymphocytes T CD8+ d'un antigène, la séquence peptidique pouvant être présentée sur une molécule de Classe I du CMH ; et produire un polypeptide ou un peptide comprenant une séquence peptidique modifiée de l'épitope des lymphocytes T CD8+ ou une molécule d'acide nucléique codant pour ledit polypeptide ou ledit peptide. La séquence peptidique modifiée porte une mutation, par rapport au peptide parent, la mutation introduisant un résidu acide aminé cyclique ou pseudo-cyclique à la position 3 de la séquence peptidique modifiée, définie de telle façon que les résidus sont numérotés séquentiellement, la position 1 correspondant au premier résidu présenté dans la fente de liaison au peptide de la molécule de Classe I du CMH, et le peptide modifié ayant une affinité de liaison plus élevée pour la molécule de Classe I du CMH et/ou générant une réponse en lymphocytes T CD8+ plus élevée, par rapport au peptide parent.
