Abstract
The present invention relates to a novel health food comprising chlorella extract, a herbal extract obtained from a mixture of herbs selected from Panax ginseng, Ganoderma lucidum, Valeriana frauriei birq, Poria cocus, Zerumbet Bupleurum scorzoneraefolium, Glycyrrhiza and along with a malt and honey. The health food has the enhancement effects of anti-oxidant, immunoenhancing, autonerve-modulating and hematopoiesis.
Description
COMPOSITION CONTAINING CHLORELLA AND HERBAL
Field of the Invention
The present invention relates to a novel health food comprising chlorella extract, a herbal extract obtained from a mixture of herbs selected from Panax ginseng, Ganoderma lucidum, Valeriana frauriei birq, Poria cocus, Zerumbet, Bupleurum scorzoneraefolium, Glycyrrhiza and along with a malt and honey. The novel health food has the enhancement effects of anti-oxidant, immunoenhancing, autonerve-modulating and hematopoiesis.
Background of the Invention
Chemical drugs, and health foods comprising the immune-enhancing factors derived from plants, for improving the immune system, have been developed in many countries. It has been known that the immune defence system is activated or suppressed by endogenous or exogenous factor. It appears that immune defence system is suppressed by many factors such as the oxidative stress, the immune cells dysfunction, the imbalance of autonerve system, hematopoiesis dysfunction, malnutrition and so on.
Oxidative stress is a stress, caused by free radicals like superoxide occurred by metabolism, various diseases, radioactive rays and the like.
Free radicals are natural by-products of daily metabolism and contribute to diseases and aging process. The presence of free radicals appears to contribute to suppression of the immune system and cells damage. Especially, it appears that the free radicals can suppress the immune cell, or macrophage, which plays essential and pivotal role in immune response. Antioxidants can counter the effects of free radicals, thereby enhancing immune function.
The amount of lymphocytes are reduced in humans with immunosupression disorders or likely diseases, and the cytokines are also reduced accordingly. Cytokines play the important role in the immune system.
Some of studies have also indicated that the immune system is related closely to the autonomic nerve system. It appears that, during stress, whether inflicted on the body through heavy exercise, severe illness or a viral infection, the sympathetic nerve system is
excited while the parasympathetic nerve system is suppressed, thereby symptoms such as insomnia, sensibilities and digestive trouble may occur. The continual statue may cause decreasing the immune defence function.
It has been considered that the inhibition of the sympathetic nervous system may be used to enhance an immune response when the endogenous immune response is not sufficient, while sthenia of the parasympathetic nervous system may strengthen immune response. The immune defence function is enhanced by the immune stimulative, hematopoiesis accelarator and nutrition.
Various drugs have a good effect in human' immune system, but the herbal extract which controls anti-oxidant action, immunocell stimulation, enhance of hematopoiesis function and autonomic nerve system is not known.
The major object of the invention is to provide health food that enhances immune defense function to treat, or prevent various diseases.
The immunomodulators, hematopoiesis-related agents and nutritional supplement enhance immune defences.
The health food according to the present invention is prepared for the reduction of oxidation stress, activation of immunocyte, the control of autonomic nerve system and the enhance of hematopoiesis system.
The health food according to the present invention comprises Chlorella extract, herbal extract obtained from a mixture of herbs selected from Panax ginseng, Ganoderma Lucidum, Valeriana fauriei briq, Poria cocus, Zerumbet, Bupleurum scorzoneraefolium, Glycyrrhiza and along with malt and honey.
Brief Description of the invention
The chlorella, called hyperaliment, is a single cell green algae, which contains polysaccharide, polysaccharide mixture and active glucan and is used as immune modulation agent. The chlorella is also rich in vitamins, mineral, esculent fiber, nucleic acids and amino acids.
Panax ginseng enhances immune response by stimulating immune cells which secretes cytocine. It promotes fatigue and sanguification preventing hemolyzation. It also has effect on sthenia, exiting systema nervosum, cardiac, sexual stimulation, digestion and absortion, as well as anti-allergy. It also has antioxidative effect.

Detailed description of the invention.
The present invention provides a health food comprising of: Chlorella extract, herbal extract obtained from amixture of herbs selected from Panax ginseng, Ganoderma Lucidum, Valeriana fauriei briq, Poria cocus, Zerumbet, Bupleurum scorzoneraefolium, Glycyrrhiza and along with malt and honey.
In an embodiment of the present invention, the content of chlorella extract is in the range about 5 mg to 50 mg, preferably 10 mg to 20 mg.
In an embodiment of the present invention, Panax ginseng is in the range about 14% to 17% of the health food.
In an embodiment of the present invention, Ganoderma Lucidum is in the range about 13% to 17% of the health food.
In an embodiment of the present invention, Valeriana fauriei briq is in the range about 12% to 18% of the health food.
In an embodiment of the present invention, Poria cocus is in the range about 10% to 15% of the health food.
In an embodiment of the present invention, Zerumbet is in the range about 10% to 13% of the health food.
In an embodiment of the present invention, Bupleurum scorzoneraefolium is in the range about 5% to 9% of the health food.

In one or more embodiment of the present invention, the percentage ratio of the malt is in the range of 8 to 10.
In another embodiment of the present invention, the health food provides antioxidant, autonerve-modulating, immune enhancing, and hematopoiesis properties.
The present invention also provides a process for preparing the health food which has antioxidant, autonerve-modulating, immune-enhancing, and hematopoiesis properties.
A detailed description of the present invention is provided in the Example section.
Example 1. Preparing of the composition according to the present Invention ,-,.;.
Preparing of the aqueous composition of the present invention. Chloralla extract; Dissolve the dried powder of chlorella in ethyl acetate, and extract at 450C for 10 hr. Centrifuge (a)solution to obtain the supernant.
Add the ethanol (80% final ethanol content)into the supernant precipitate the active agents. Filter the precipitate, wash with absolute ethanol and dry the extracts under vacuum.
Herbs extract;
Mix and crush the Panax ginseng, Ganoderma lucidum, Valeriana fauriei briq, poria cocus,
Zerumbet, Bupleurum scorzoneraefolium, Glycyrrhiza with size of about 0.001-10mm3.
Add the crushed mixture into the distilled water, or ethanol and extract for 5h at 8O0C.
Centrifuge extracted solutions to obtain the supernant.
Malt;
Steam the rice bran, and then inoculate with Aspergillus oryzae.
Ferment at 20°C-25°C for 2 to 3 days,
Self-degradation at 50°C-80°C for about 10 hours, Add the distilled water into the Self -degraded solution,
Centrifiige the mixed solution to obtain the supernatants.
Mixture; chlorella extracts, malt extracts and herbs extracts obtained from above steps mix and add the honey to composition of the present invention.
2. Preparing of the solid composition of the present invention.
1) Mix and crush the Panax ginseng, Ganoderma lucidum, Valeriana fauriei briq, poria cocus, Zerumbet, Bupleurum scorzoneraefolium, Glycyrrhiza,.
2) Add the crushed mixture into the distilled water, extract for 24h at 6O0C.
3) Extracted solution filter and concentrate,
4) mix the concentrated materials with the chlorella extracts and malt extracts and then malaxate with the suitable honey,
5) prepare the pill with the malaxated mixture
As above process, the compositions of the present invention and contents their compenents are shown tables 1, 2, 3 and.

Example 2. Measurement of anti-oxidant activity
Since elimination of DPPH free radicals changes the color of DPPH solution, anti-oxidant activity was measured, as follows;
DPPH of 5mg was dissolved in 100ml of 60% methanol to prepare a DPPH solution. Sample solution; mixed solution of the DPPH solution (750/^1) and the UC(2Oθ£tl). Blank solution; mixed solution of 60% methanol (750 μ.1) and UC solution (200 ^tI).
Control solution; mixed solution of the DPPH solution (750 μ, 1) and 60% methanol(200/xl).
Absorbance of each solution was measured at 515nm. Anti-oxidant activity (elimination activity of free radicals) was calculated from the following formula;

The above procedure was applied to invention composition 1, 2, 3 or 4 respectively. The antioxidant activities are shown in the following Table 5.

As shown in the above table, the present invention compositions showed very high antioxidant activity.
Example 3. Measurement of hematopoiesis function
Radiation destroys bone marrow cells and has a negative effect on the generation and replication of normal immune cells, thereby deteriorating the function of hematopoiesis and immunization. In the present experiment, it has examined whether the administration of UC can reduce the deterioration of hematopoiesis and immune function accompanying radiation treatment.
After the mice weighing 24-27g were exposed to 2Gy of 60Co, gamma, 0.3ml/kg of UC 2 was orally administered to mice for 30 days. 10 days, 20 days and 3Od ays after irradiation, irrespectively, the mice were killed to harvest the peripheral blood to measure the number of erythrocyte(red blood cell:RBC), leucocytes (white blood celkWBC) and blood ρlatelet(BP).

Example 4. Effect on the modulation of the autonerve system.
hi the present experiment, the effect of UC 1 to modulate the autonerve system is measured by the method of heart rate variability-check. ,
The patients are laid down on the bed for 5min to lOmin and then conductor electrocardiogram II (ECG II ) is set up on the patients's limb. The electrocardiogram data is input continuously to computer for 5min.
The ratios of LF/HF is reflected the function of sympathetic nerve system.
0.3ml/kg of the UC 2 were orally administrated to patients with insomnia, 2h after administration; LF/HF was measured according to above method.

As shown in the above table the present invention composition showed the effect on the invention of sympathetic nerve system and sthenia of the parasympathe tic nerve system.
Example5. Effect of the invention composition on phagocytosis of cells from peripheral blood.
A group of mice has been injected intraperitonealy with with PBS(control) or UC.
24 later, mice were scarified, peripheral blood from the orbital plexus was collected into heparine(5iv/ml).
After counting, a test of phagocytosis of yeast partides was performed.
0.1ml of heparinzed fresh blood was add to 0.05ml of diluted yeast particles(5*108/ml) and incubated for 60 minutes at 370C with occasional gentle agitation.
After the end of incubation, the cell suspension was smeared over microscope slides.
Smears were evaluated under the optical microscope after Accustain staining.
Cells with at least three engulfed particles were considered to be positive.
The results are given in the following tableό. the results clearly show that the invention composition strongly stimulate phagocytosis in both monocytes and granulocytes.

Example 10. Effect of the invention composition on the engulfment of cells from peritoneal cavity
A group of mice has been injected intrapritonealy with PBS(control) or UC. 24 later, mice were scarified, peritoneal cells were collected into medium. After counting the cells in hemocytometer, the peritoneal cells were dilated to l*107cells in medium with 5% fetal calf serum.
2* 106 cells in 0.2ml of medium supplemented with 5% fetal calf serum were mixed with the same volume of yeast particles(5*108/ml). The suspension was incubated for 60 minutes at 370C with occasional agitation. Incubation was terminated by centrifugation (150g for 5minute) and the pellet was resupended.
Macrophages with ingested particles were scored under the optical microscope in the smears after. Accustain staining. Cells with four or more engulfed particles were considered to be positive.
The results are given in the following table7. they show that the invention composition strongly stimulate phagocytosis of peritoneal macrophages.

1. A composition comprising of: a chlorella extract, a concentrated herbal extract obtained from a mixture of herbs selected from Panax ginseng, Ganoderma lucidum, Valeriana fauriei briq, Poria cocus, Zerumbet Bupleurum scorzoneraefolium, Glycyrrhiza and malt extract along with honey.
2. The composition food as claimed in claim 1, which comprises chlorella extract in an amount of about 5 mg to 50 mg, preferably 10 mg to 20 mg; Panax ginseng in an amount of about 14% to 17% , Ganoderma Lucidum in an amount of about 13% to 17% , Valeriana fauriei briq in an amount of about 12% to 18%, Poria cocus in an amount about 10% to 15%, Zerumbet in an amount of about 10% to 13%, Bupleurum scorzoneraefolium in an amount of about 5% to 9%, Glycyrrhiza in the amount of about 6% to 7% and the malt extracts in the amount of about 8% to 10% of the health food.
3. The composition as claimed in claim 1, in which the malt extracts are obtained by inoculating with Aspergillus oryzae in the agent for malt such as the rice bran.
4.The composition claimed in any of claim 1 to 2, which has a tablet or a solution.
5. Use of composition claimed in any of claims 1 to 3, in which the composition is used as the health food having anti-oxidant, hematopoiesis-enhancing, autonerve-modulating and immuno-enhancing properties
6. The said health food as claimed in claim 5, which has anti-oxidant activity.
7. The said health food as claimed in claim 5, which has the activity of hematopoiesis- enhancment.
8. The said health food as claimed in claim 5, which has the effect on the relaxation of sympathetic nerve system and/or the sthenia of parasympathetic nerve system.
9.The said health food as claimed in claim 5, which has the immune defence function against radiation.
