Abstract

[PROBLEMS] To provide: a method for the determination of a high-density lipoprotein (HDL) cholesterol level in a biological sample such as a serum and a plasma; and a reagent for use in the method. [MEANS FOR SOLVING PROBLEMS] Disclosed is a method for the determination of a cholesterol level in a high-density lipoprotein by an enzymatic reaction, which comprises: a first reaction step for preferentially scavenging cholesterol in a low-density lipoprotein, a very-low-density lipoprotein and chylomicron contained in a sample, wherein the first reaction is carried out in a buffer solution containing a cholesterol esterase, a cholesterol oxidase and a substance capable of inhibiting the reactivities of the enzymes against the high-density lipoprotein; and a second reaction step for determining the cholesterol level in the high-density lipoprotein, wherein the second reaction is carried out in a buffer solution which is the same as the buffer solution used in the first reaction step except that a substance capable of reducing or eliminating the inhibitory activities of the enzymes against cholesterol in the high-density lipoprotein in the first reaction is additionally contained.
