Abstract

L'invention concerne des articles et des procédés permettant de déterminer un analyte indiquant une condition de maladie. Dans certains modes de réalisation, les articles et procédés décrits ici peuvent être utilisés pour déterminer, qualitativement ou quantitativement, une présence d'un composant, tel qu'un type de cellule particulier, dans un échantillon de fluide. Dans un mode de réalisation particulier, on propose un système microfluidique à faible coût permettant une détection rapide de cellules T. Le système microfluidique peut utiliser des anticorps immobilisés et des molécules d'adhérence dans un canal pour capturer les cellules T à partir d'un échantillon de fluide tel qu'un petit volume de sang. Les cellules T capturées peuvent être étiquetées avec un colloïde métallique (par exemple des nanoparticules d'or) en utilisant un anticorps spécifique au récepteur de cellules T (TCR), et de l'argent métallique peut être précipité de manière catalytique sur les cellules. Le nombre de cellules T capturées peut être compté et peut indiquer une condition de maladie d'un patient telle qu'une déficience immune combinée sévère ou un virus d'immunodéficience humaine.
