Abstract

L'invention concerne des procédés améliorés d'apport transdermique de formulations médicamenteuses; des formulations conçues pour améliorer l'apport, dans la circulation systémique, de taux thérapeutiques de médicaments appliqués par voie topique; des procédés de fabrication des formulations. Les formulations contiennent au moins un agent actif à administrer et au moins un agent destiné à améliorer la perméabilité de la peau dans un hydrogel polymère, et des excipients supplémentaires facultatifs. L'invention concerne des procédés permettant d'améliorer l'apport de formulations dans et à travers la peau, qui comprennent les étapes consistant à: (a) prétraiter la peau à l'aide d'une solution hydratante, (b) rendre la couche cornée physiquement perméable par l'application d'ultrasons à basse fréquence (administrés par un dispositif de sonophorèse tel que le SONOPREP®, vendu par Sontra Medical Corporation, Franklin, MA), (c) appliquer par voie topique une formulation contenant la molécule bioactive, et éventuellement (d) appliquer une différence de potentiel électrique qui force les médicaments ionisés à traverser la peau. La formulation contient éventuellement des agents améliorant la perméabilité. Le procédé s'utilise avec les formulations décrites ou avec d'autres formulations s'administrant par voie topique. Dans un mode de réalisation préféré, l'agent actif à administrer est un médicament. De préférence, le médicament est un anesthésiant local tel que la lidocaïne.
