Abstract

L'invention concerne un procédé destiné à la détection d'un agent pathogène dans un échantillon. Ce procédé consiste à mettre un lysat de cet échantillon en contact avec une sonde de capture immobilisée sur un substrat. Ce lysat peut être préparé rapidement à partir d'un échantillon clinique, ce qui évite le recours à une culture d'échantillons ou à une épuration bactérienne avant l'analyse. La sonde de capture comprend un premier oligonucléotide qui s'hybride spécifiquement avec une première séquence nucléotidique de l'agent pathogène à détecter. Le lysat est mis en contact avec une sonde de détection comprenant un oligonucléotide marqué, décelable, qui s'hybride spécifiquement avec une seconde séquence nucléotidique de l'agent pathogène. La détection du second marqueur décelable combiné avec le substrat indique la présence de l'agent correspondant. L'invention concerne également des variations de ce procédé, permettant d'optimiser la détection. L'invention concerne également des sondes et des trousses destinées à être utilisées dans ce procédé.
