Abstract

La présente invention concerne essentiellement des bibliothèques combinatoires de molécules marqueurs de protéines comprenant des groupes qui peuvent être activés chimiquement et photochimiquement, avec ou sans groupes rapporteurs, reliés en des points de diversité ou selon des agencements spatiaux différents via des chaînes latérales autour d'un noyau moléculaire commun. L'invention se rapporte en outre à des bibliothèques combinatoires de motifs marqueurs comprenant des groupes marqueurs chimiquement ou photochimiquement réactifs, des groupes rapporteurs et divers autres types de chaînes latérales attachées de façon variable à un noyau structurel moléculaire, de préférence à base de lysine. L'invention concerne aussi un procédé d'arrimage chimique combinatoire selon lequel une chaîne latérale comprenant un groupe fonctionnel terminal, de préférence un groupe amine, et à laquelle un groupe marqueur est facultativement attaché, est introduite dans une position optimale garantissant la diversité et l'efficacité de la liaison à la protéine. L'invention permet de simplifier la complexité du protéome dans différents tissus et d'identifier des marqueurs de protéines destinés au diagnostic en faisant alterner différents motifs de marquage dans des tissus sains et malades.
