Abstract

Selon l'invention, une zone locale transversale, une charge locale ou la chimie locale d'une molécule est modifiée en vue de sonder les caractéristiques d'une molécule d'acide nucléique cible ou d'une population de molécules d'acide nucléique cibles. L'acide nucléique modifié est mis en contact avec un substrat comprenant un détecteur répondant à la modification dans la zone locale transversale, dans la charge locale ou dans la chimie locale de la molécule d'acide nucléique. La molécule d'acide nucléique modifiée traverse un volume défini et, de préférence, aux dimensions moléculaires, sur le substrat, de manière que des nucléotides de la molécule d'acide nucléique modifiée interagissent avec le détecteur dans un ordre séquentiel, des données en corrélation avec la zone transversale, la charge locale ou la chimie locale de la molécule d'acide nucléique étant obtenues. La molécule d'acide nucléique peut être modifiée au niveau de plusieurs emplacements situés le long de la longueur de la molécule. L'interaction des nucléotides individuels de la molécule d'acide nucléique avec le détecteur dans un ordre séquentiel permet d'obtenir des informations relatives à l'emplacement et à la composition d'une pluralité de sites modifiés le long d'une molécule unique.
