Abstract

L'invention concerne des procédés de détermination d'un ou plusieurs effets biologiques d'un composé sur l'expression génétique. Ces procédés consistent à obtenir un extrait nucléaire de cellules exposé à un composé, puis à combiner cet extrait avec un acide nucléique, ou une librairie d'acides nucléiques, contenant un ou plusieurs éléments régulateurs dans des conditions permettant la formation de complexes cis/trans entre les éléments régulateurs et les composants (par ex., les protéines de liaison d'ADN) de l'extrait nucléaire. Les complexes ainsi formés sont comparés aux complexes formés au moyen des extraits nucléaires obtenus de la même manière, mais dont les cellules n'ont pas été traitées, et au composé. Les différences entre les complexes formés après exposition des cellules au composé peuvent ensuite être évaluées. Les procédés de l'invention sont, de préférence, exécutés dans une configuration à débit élevé, et sont utiles, par exemple, afin d'évaluer l'efficacité, la toxicité, et le mécanisme d'action d'un composé. Par ailleurs, cette invention sera utile dans la mise au point de nouveaux médicaments, et dans la meilleure compréhension et amélioration de composés déjà utilisés ou en cours de mise au point.
