Abstract

Cette invention concerne des procédés, des systèmes et des dispositifs permettant d'analyser un matériau biologique, tel qu'un spécimen cellulaire ou autre. Selon un mode de réalisation, la méthode consiste à transférer sélectivement et focalement des biomolécules d'une région cible d'intérêt à un échantillon biologique (tel qu'un fragment de tissu). Pour le transfert, on peut, par exemple, modifier sélectivement une couche de transfert adjacente à la région cible de sorte que la biomolécule puisse traverser la zone modifiée de la couche de transfert. Dans des exemples particuliers, on modifie la couche de transfert en en augmentant focalement la perméabilité, par exemple en retirant une partie focale de la couche de transfert et en transportant des biomolécules au travers de la région modifiée de manière à pouvoir séparer par microdissection des molécules d'intérêt de l'échantillon biologique. Selon d'autres modes de réalisation, les biomolécules soumises à microdissection peuvent être appliquées sur un substrat d'analyse renfermant une molécule d'identification, tel qu'un jeu d'acides nucléiques, un champ de balayage d'expression en couche, ou des puits renfermant des anticorps. La microdissection de transfert permet d'analyser sélectivement des biomolécules extraites de régions d'intérêt. Il est par exemple possible de déterminer l'expression différentielle de certaines protéines par l'analyse de nids de cellules fortement atypiques ou métastatiques dans une partie de tumeur.
