Abstract

L'invention concerne une graine d'arachide transgénique non allergène obtenue par des techniques de recombinaison. Les plantes d'arachide sont transformées à l'aide de multiples copies de chacun des gènes allergènes, ou de leurs fragments, afin de supprimer l'expression génique et la production de protéines allergènes. En variante, les plantes d'arachide sont transformées à l'aide de gènes d'arachide allergènes anti-sens introduits dans le génome de l'arachide comme fragments anti-sens, fragments sens, ou des combinaisons de fragments anti-sens et sens. Les transgènes d'arachide sont régulés par le promoteur 35S ou le promoteur du gène Ara h2 pour produire des ARNs anti-sens, des ARNs sens, et des ARNs double-brin afin de supprimer la production de protéines allergènes dans les plantes d'arachide. Un clone génomique pour l'allergène Ara h2 est isolé et séquencé. L'ORF est de 622 nucléotides. La protéine codée prévue est de 207 acides aminés et comprend un peptide de transit reconnu de 21 résidus. Un signal de polyadénylation est identifié sur la position 951. Six codons d'arrêt supplémentaires sont observés. Une région promoteur s'est avérée contenir une boîte TATA reconnue située sur la position -72. Des régions homologues ont été identifiées entre Ara h2, h6, et h7, et entre Ara h3 et h4, et entre Ara h1P41B et Ara h1P17. Les régions homologues sont utilisées dans le criblage de la bibliothèque génomique de l'arachide afin d'isoler tous les gènes allergènes de l'arachide et pour la régulation négative et l'inhibition des multiples gènes allergènes de l'arachide.
