  • the mixture may be directly heated, since it is quite difficult to appropriately control the temperature, it is preferred that an airtight container containing the mixture of sodium chloride solution and dog sperm is submerged in hot water at about 50° C and heated.

  • a mixture having the following composition was homogeneously mixed using a mixer to prepare the composition for freezing dog sperm based on 100 ml of distilled water.

  • the present invention provides a method for freezing dog sperm which is difficult to be frozen for storage in view of peculiarity of spermatozoa of the dog sperm, and compositions for use therein. Therefore, the dog sperm can be stored in a frozen state, thereby preserving the dog of good strain. Also, the frozen male dog sperm can be used for artificial insemination at an appropriate time and place, thereby enabling an increase in the birth rate.

Abstract

L'invention concerne des compositions permettant de congeler du sperme canin, un procédé de congélation de sperme canin utilisant ces compositions et un procédé d'insémination artificielle employant le sperme canin congelé. La composition permettant de congeler du sperme canin renferme 0,25 à 4,5g de citrate de sodium, 0,45 à 4,0g de dextrose, 0,01 à 0,3g de pénicilline, 50 000 à 500 000 UI de streptomycine, 0,25 à 0,45g de catalase, 0,045 à 0,25g de jaune, 1,5 à 15ml de glycérol, et 0,02 à 0,1 ν)g de sérum canin. Le procédé de congélation de sperme canin de cette invention consiste tout d'abord a) à recueillir du sperme d'un chien mâle par massage ou application d'une décharge électrique, b) à centrifuger le sperme recueilli de manière à séparer le fluide et à fractionner le sperme resté au fond du tube, c) à mélanger ce sperme resté au fond du tube à la composition susmentionnée, selon un rapport volumétrique de 1:1, puis à réfrigérer ce mélange une première fois, d) à ajouter de nouveau ladite composition au mélange obtenu au cours de l'étape précédente, de sorte que le sperme et la composition peuvent se mélanger selon un rapport volumétrique de 1:2, ce mélange étant ensuite réfrigéré une seconde fois, et enfin e) à ajouter de l'azote liquide au mélange obtenu au cours de l'étape précédente avant de congeler celui-ci.
