Abstract

Die Erfindung betrifft Verfahren zur gleichzeitigen Identifizierung eines Proteins und seines Bindungspartners, das dadurch gekennzeichnet ist, daß man a) Proteine oder Proteinaggregate aus einer biologischen Quelle isoliert und auftrennt; b) die aufgetrennten Proteine oder Proteinaggregate auf einer Oberfläche immobilisiert; c) eine kombinatorische Bank mit den auf einer Oberfläche immobilisierten Proteinen oder Proteinaggregaten inkubiert; d) diejenigen Mitglieder der kombinatorischen Bank, die an die immobilisierten Proteine binden, von nicht-gebundenen Mitgliedern der Bank trennt; e) die an die Oberfläche gebundenen Komplexe aus Protein und Bindungspartner aus der kombinatorischen Bank isoliert; f) die Proteine in den so isolierten Komplexen mit einer Kombination aus einem physikalisch-chemischen Verfahren identifiziert; und g) gegebenenfalls die isolierten Bindungspartner anreichert. Die vorliegende Erfindung erlaubt somit die gleichzeitige Identifizierung von Proteinen mit und ohne vorherige Reinigung, sowie die Auswahl von Mitgliedern kombinatorischer Banken, die mit diesen Proteinen wechselwirken. Dadurch läßt sich auf einfache Weise die Funktion der Proteine über ihre spezifischen Bindungspartner ermitteln.
