Abstract
A method for the determination of nucleic acids is disclosed wherein in a first extension reaction a multiplicity of extension products of a first set of primers is produced, said extension products being digestible by nuclease, and in a second extension reaction extension products of a second set of primers are produced using said first extension products as templates, wherein said primers of said second set are not capable of serving as a template for the extension of a complementary primer. Intersample cross contamination can be reduced by subjecting said first extension products to nuclease treatment.
