Abstract

Eine ausgezogene Faserspitze (1) wird mit Photobiotin belegt und mit UV-Licht bestrahlt. Dadurch geht das Photobiotin mit der Faserspitze in Bindung. Die Faserspitze wird anschließend mit Streptavidin in Kontakt gebracht und in eine Lösung getaucht, die gefärbte Kopien des zu sequenzierenden DNA-Moleküls enthält. Am 5'-Ende der gefärbten DNA-Moleküle (4A) ist ebenfalls Biotin gebunden. Das an die einzelnen DNA-Moleküle gekoppelte Biotin bindet an das Streptavidin an der Faserspitze. Zum Nachweis dieser Bindung wird Licht in die Faser eingekoppelt. Das evaneszente Feld dieses Lichts regt ein gefärbtes DNA-Molekül zur Fluoreszenz an, sobald es an die Faserspitze bindet. Wird eine Bindung festgestellt, wird die Faser sofort aus der Lösung entfernt. Die Faserspitze wird sodann in eine Mikrokapillare (5) eingeführt. Die Mikrokapillare wird mit einer Pufferlösung gefüllt, die Exonukleasen enthält, die das DNA-Molekül Base für Base abbauen. Der Ausgang (6) der Kapillare befindet sich im Beobachtungsbereich einer Einzelmolekül-Detekionsapparatur. Um das Signal-Rauschen-Verhältnis für die Einzel-molekül-Detektion zu verbessern, ist die Wandstärke der Mikrokapillare im Bereich der Austrittsöffnung deutlich kleiner als 1/4 der Wellenlänge des Anregungslichts.
