Abstract
A composition for delaying release of an active component into an aqueous fluid is taught which includes a capsule that swells in the presence of an aqueous fluid allowing moisture to enter the capsule into contact with a capsule decomposing agent activated by the entering moisture such that the agent decomposes the capsule sufficiently to release the active agent at a predetermined delayed time after the capsule contacts the aqueous fluid. A preferred capsule composition is gelatin and decomposing agents are a proteolytic enzyme or a salt that affects the osmotic gradient of the system. The controlled release composition of the invention is preferably utilized in a soft contact lens disinfecting system wherein a solution of hydrogen peroxide disinfects the lenses followed by neutralization of residual hydrogen peroxide wherein release of the neutralizing agent is delayed by means of the capsule proteolytic enzyme or salt composition.
Claims
1. A composition for delaying release of a neutralizing composition into a solution of hydrogen peroxide for disinfecting contact lenses, comprising:
a gelatin capsule that swells in said solution allowing moisture to enter said capsule; a capsule decomposing agent that is a proteolytic enzyme or a water soluble inorganic or organic salt that is activated by said entering moisture wherein said agent decomposes said capsule sufficient to release said neutralizing component into said solution, wherein release of the neutralizer is delayed until after the disinfecting step is completed and then neutralize residual hydrogen peroxide.
2. The composition of claim 1 wherein said neutralizer component further comprises a proteolytic enzyme within said capsule wherein the capsule in the aqueous medium swells, transmitting sufficient water into the capsule to activate the proteolytic enzyme to digest the capsule, releasing the neutralizer into the aqueous medium whereby residual H2O2 is neutralized.
3. The composition of claim 2 wherein said proteolytic enzyme is subtilisin.
4. The composition of claim 2 wherein said neutralizer is a catalase enzyme.
5. The composition of claim 10 wherein the amount of subtilisin is adjusted to delay digestion of said capsule to provide sufficient time for disinfecting by said H2O2 solution.
6. The composition of claim 5 wherein said subtilisin is present in the amount of about 0.05-3.00 milligrams per capsule and said catalase release is substantially delayed for about 1-2 hours.
7. The composition of claim 1 wherein said means for delaying release of said catalase enzyme comprises sodium chloride within said capsule.
8. The composition of claim 7 wherein said NaCl is present in said capsule in a range of about 5-100 milligrams per capsule.
9. The composition of claim 7 wherein said NaCl is present in less than about 70 milligrams per capsule.
10. The composition of claim 7 wherein said NaCl is about 10 milligrams per capsule and said catalase release is substantially delayed for about 2-3 hours.
11. A process for delaying release of a neutralizing composition into a solution of hydrogen peroxide for disinfecting contact lenses, comprising: placing a gelatin capsule that swells in said solution and allowing moisture to enter said capsule; said gelatin capsule containing a decomposing agent that is a proteolytic enzyme or a water soluble inorganic or organic salt that is activated by said entering moisture wherein said agent decomposes said capsule sufficient to release said neutralizing component into said solution, wherein release of the neutralizer is delayed until after the disinfecting step is completed and then neutralize residual hydrogen peroxide.
12. The process of claim 11 wherein said neutralizer component further comprises a proteolytic enzyme within said capsule wherein the capsule in the aqueous medium swells, transmitting sufficient water into the capsule to activate the proteolytic enzyme to digest the capsule, releasing the neutralizer into the aqueous medium whereby residual H2O2 is neutralized.
13. The process of claim 12 wherein said proteolytic enzyme is subtilisin.
14. The process of claim 12 wherein said .neutralizer is a catalase enzyme.
15. The process of claim 14 wherein the amount of subtilisin is adjusted to delay digestion of said capsule to provide sufficient time for disinfecting by said H2O2 solution.
16. The process of claim 15 wherein said subtilisin is present in the amount of about 0.05-3.00 milligrams per capsule and said catalase release is substantially delayed for aobut 1-2 hours.
17. The process of claim 11 wherein said means for delaying release of said neutralizer comprises sodium chloride within said capsule.
18. The process of claim 17 wherein said NaCl is present in said capsule in a range of about 5-100 milligrams per capsule.
19. The process of claim 17 wherein said NaCl is present in less than about 70 milligrams per capsule.
20. The process of claim 17 wherein said NaCl is about 10 milligrams per capsule and said catalase release is substantially delayed for about 2-3 hours.

STATEMENT UNDER ARTICLE19
This Statement accompanies the amendments under PCT Article 19 submitted herewith, wherein original claims 1-28 are replaced by amended claims 1-20.
Original claims 1-5, 7 and 17-28 have been canceled. Original claim 6, now claim 1, has been amended to indicate that the capsule is a gelatin capsule and that the decomposing agent within the gelatin capsule is a proteolytic enzyme or a water soluble inorganic or organic salt. Support for the term "gelatin" is found on page 7 of the Specification, line 7 and 8 thereof. Support for the amendment relating to the decomposing agent is found on page 7 of the Specification, lines 10-15 thereof. New claims 2-10 correspond to original claims 8-16, respectively, with the dependencies corrected to reflect the renumbering of the new claims. New claims 7 and 10 have also been amended by deleting terms "catalase" and "catalase enzyme" and replacing them with the term "neutralizer." There is no antecedent basis in independent claim 1 for the term "catalase," while there is support for the term "neutralizer."
Applicant has presented new claims 11-18. These claims are all process claims and they track new composition claims 1-10.
The claims, as amended, of related case PCT/US93/12553 (filed 21/12/93) are directed to a preserved saline solution for disinfecting and cleaning contact lenses comprising a solid PVP- H202 hydrogen peroxide complex and a solid neutralizing component for neutralizing H202 wherein the neutralizer component comprises a gelatin capsule containing a neutralizing component and a capsule decomposing agent. The claims for that case also include a method for disinfecting and cleaning contact lenses using the foregoing preserved saline solution containing solid PVP-H202 complex and a gelatin capsule containing a neutralizing component and a decomposing agent. The claims of the present PCT application are directed to a method and composition for delaying release of a neutralizing composition into a solution of hydrogen peroxide for disinfecting contact lenses. In other words, the related case is directed to a solid PVP-H202 composition whereas the present application is directed to a solution of hydrogen peroxide.
