Abstract

La invención se refiere a un proceso para la producción de material vegetal genéticamente modificado que comprende una o más secuencias de ADN de interés establemente incorporadas, comprendiendo tal proceso someter material vegetal que consiste de, o comprende, brotes, extremos o ápices de brotes, brotes enraizados o plántulas a una transferencia de la(s) secuencia(s) de ADN mediante Agrobacterium, siendo dicha transferencia de ADN mediante Agrobacterium llevada a cabo en un medio de cultivo líquido oxigenado, por ejemplo aireado, en el cual el material vegetal es cultivado, estando el material vegetal sumergido en el medio líquido. El medio generalmente es agitado o movido de otro modo. El material vegetal preferentemente es cultivado activamente en el medio de cultivo sumergido. El medio líquido está mantenido y con un pH determinado de modo tal que el crecimiento vegetal procede normalmente, pero el crecimiento del bacterium está restringido y a su vez que estimule la transferencia del ADN de interés desde el bacterium al material vegetal. Debe comprender suficiente oxígeno para soportar el metabolismo del material vegetal. Generalmente es necesario proveer oxígeno, usualmente en la forma de aire, y/o iluminar el sistema de modo que se produce oxígeno por fotosíntesis. También debe proveerse una fuente de carbono apropiada, por ejemplo, dióxido de carbono o una fuente de carbono orgánico. El material vegetal es obtenido a partir de un cultivar, clon o siembra. Puede ser una planta anual, bianual o perenne; planta mono o dicotiledónea; planta herbácea; planta leñosa, etc. La formación de brotes es inducida en el material vegetal genéticamente modificado en presencia de un agente capaz de inducir la formación de brotes, pudiendo ser este agente inductor una citoquina, 6-bencilaminopurina ó N-(2-cloro-4-piridil)-N'-fenilurea.
