在實施例中,相較於不含生物活性玻璃(亦即,不存在生物活性玻璃)的牙科配方基底配方,接觸牙科配方的步驟具有50至95%的閉塞暴露的牙本質小管的較高相對速率。
在實施例中,接觸牙科配方的步驟係可選自下列至少一者:以糊狀物配方研磨、以液體配方沖洗、注入液體配方、以複合材料(例如,通常由粉末化生物活性玻璃與丙烯酸樹脂製得)填充牙齒、或前述步驟的組合。
在實施例中,牙齒表面可具有下列至少一者,例如:牙本質表面、牙本質小管或牙本質表面與牙本質小管的組合。

在實施例中,本發明在數種態樣中是有利的,例如包含:所揭露之硼酸鹽與硼矽酸鹽玻璃組成物可具有比含45S5牙膏明顯較高的閉塞暴露牙本質小管的速率,此結果展現所揭露之生物活性玻璃對於治療牙本質高敏性或牙本質敏感性的優良性質;所揭露之生物活性玻璃組成物可不含無機鉀鹽,且所揭露之生物活性玻璃組成物與牙本質治療配方可不含矽酸鹽。Novamin®與Biomin®為含矽酸鹽的生物活性玻璃。
在實施例中,本發明提供硼酸鹽與硼矽酸鹽生物活性玻璃組成物,例如,如表1所示,該硼酸鹽與硼矽酸鹽生物活性玻璃組成物可與哺乳類的唾液反應並釋放鈣與磷酸鹽,以形成碳酸羥基磷灰石、或假如存在氟化物離子,會形成氟磷灰石(請參閱第2圖與第3圖),藉由在牙本質小管中形成塗抹層展現表面處的小管閉塞,且重建、強化並保護牙齒結構。亦在磷灰石層中沉積有微量鎂(第3圖)。正常的牙釉質與牙本質分別包含,例如,0.44與1.23重量%的鎂。鎂是許多酶的輔助因子,且穩定DNA與RNA的結構。鎂對於牙齒的成長與維護可具有刺激作用。表 1 生物活性玻璃組成物(莫耳百分比)
可將氟化物以前驅物的形式併入玻璃組成物中,例如,氟化鈉(NaF)、氟化亞錫(SnF2
)、氟化鈣(CaF2
)與類似氟化物或前述氟化物的混合。併入氟化物的玻璃可釋放氟化物並在唾液中形成氟磷灰石。氟磷灰石比碳酸羥基磷灰石更能抵抗酸溶解。
亦可將併入氟化物的玻璃添加至非水溶性的潔牙劑介質,例如,與其他生物活性玻璃組合。已知氟化物離子可幫助磷灰石的形成且可刺激造骨細胞分裂。
所揭露的玻璃組成物與配方除了再礦化牙膏功效之外,所揭露的玻璃組成物與配方亦可抗菌,此性質准許,例如,避免蛀牙、降低牙菌斑生成、控制牙齦炎、抑制呼吸惡臭與類似有益的效果。所揭露的玻璃組成物與配方亦可併入其他專業應用的牙科產品,例如,清潔與研磨膏、清漆、再礦化填充材料與類似配方。
在實施例中,本發明提供再礦化組成物,包含,例如: 有效量(例如,0.1至10重量%)的生物活性玻璃組成物,包含以下來源(基於該組成物總量為100莫耳%): 2至70%的B2
O3
, 1至60%SiO2
, 2至16%的Al2
O3
, 5至30%的CaO, 1至6%的P2
O5
, 1至20%的Na2
O, 1至20%的K2
O,以及 1至20%的MgO;以及可選擇性進一步包含適合載體,基於該組成物與載體的總重量,適合載體為80至99.9重量%,生物活性玻璃為0.1至20重量%。基於該組成物總量為100重量%,來源的莫耳%具有下列等值的重量百分比:1至60%的B2

參照圖式,第1圖顯示所揭露生物活性組成物的化學與物理作用的假設方案。儘管未受理論限制,相信作用機制包含,例如:生物活性玻璃顆粒與唾液或水反應,以將離子,例如,Na+
、K+
以及類似離子與H+
交換,並提高pH(100);鈣與磷酸鹽沉積為磷酸鈣(110);以及使磷酸鈣或氟化物(假如存在)結晶,以在牙本質表面上或牙本質小管中形成碳酸羥基磷灰石或氟磷灰石(120)。
第2A至2F圖顯示進行一週治療後的牙本質薄片測試的SEM顯微圖:利用基底牙膏配方(2A、2B);利用含45S5的牙膏(2C、2D);或含實例1的牙膏(2E、2F),並浸入人造唾液中。基底牙膏治療沒有閉塞牙本質小管。利用含45S5的牙膏治療後,結晶狀沉積物部分閉塞牙本質小管。利用含實例1的牙膏治療後,結晶狀沉積物閉塞大部分的牙本質小管與牙本質表面。
第3A至3C圖顯示以包含實例1的牙膏進行一週治療後的牙本質位置的SEM-EDS分析。第3A圖顯示的SEM具有選擇用於進一步分析的兩個點(圓圈處)。顯示形成在牙本質小管上與形成在牙本質小管中的磷灰石的SEM-EDS分析為鈣與磷酸鹽沉積物與少量鎂(參閱第3B圖(點003)與第3C圖(點004))。 材料與方法
分別將表1中的每個玻璃組成物熔融並使用,例如,噴氣研磨機,將每個玻璃組成物磨碎至1至10微米。
分別配製表1中的每個磨砂玻璃組成物為諸如表2所列之牙科配方。
評估每個牙科配方,以能夠閉塞由人類臼齒所形成的牙本質薄片,詳述於實例9中,亦即,將具有牙本質小管的薄片暴露於牙科配方且與不具有所揭露之生物活性玻璃的牙科配方基底(控制)進行比較。表 2 基底與示例性含 BG* 牙膏組成物
用於製造本發明組成物的原始材料、設備或原始材料與設備可引入某些非意圖添加且可存在於最終玻璃組成物中的雜質或成分。此材料可少量存在於所揭露之組成物中並稱為「混入材料(tramp materials)」。
所揭露之組成物可包含通常是少量的混入材料。類似地,「不含鐵」、「不含鈉」、「不含鋰」、「不含鋯」、「不含鹼土金屬」、「不含重金屬」、或類似描述代表非目的性添加混入材料至組成物中,但該組成物仍可包含近似混入或少量的鐵、鈉、鋰、鋯、鹼土金屬或重金屬等等。
除非特別指示,在此所描述的所有成分濃度以重量百分比(重量%)表示。實例
以下實例根據前述一般程序展示所揭露之組成物、配方與方法的製造、使用與分析。實例 1
玻璃 1
表1中的實例1的組成物由以下方式製備:個別合併指示量的來源批次材料,包含二氧化矽、硼酸、氧化鋁、氧化鈉、氧化鉀、石灰石、氧化鎂與磷酸鈣。於塑膠瓶中使用Turbular混合器劇烈混合來源批次材料。接著將來源批次材料移至內部容積約650cc的鉑坩鍋中。接著將坩鍋載入退火爐以在250℃下鍛燒批料達24小時。接著將經鍛燒的批料於1100至1300℃下熔融達6小時,且接著將玻璃熔體傾注在鋼板上,並在400至500℃下退火。實例 2 至 6

牙科配方
以下列方式製備表2的牙科配方:首先將正丁醇與PEG 400加入玻璃組成物中,接著將配方中的剩餘成分分別加入並以刮勺完全地混合。實例 9
牙科配方的評估
以下列方式評估表2的牙科配方:由bforbones(bforbones.com)合法取得人類臼齒。使用Buehler低速水冷鑽石鋸在牙骨質-牙釉質接面上方垂直於牙齒長軸切割出厚度1mm的牙本質薄片。使用500粒度的砂紙打磨30秒,以在牙本質薄片的兩側產生塗抹層。接著利用6%檸檬酸處理3分鐘、接著利用水沖洗並在37℃下乾燥,以接續移除塗抹層。將牙本質薄片隨機區分為控制組或實驗組,每一組包含至少3個樣品,接受非水溶性牙膏基底配方或含5%的45S5牙膏,以及含所揭露玻璃組成物(如表1與表2所示)的牙膏。使用牙膏與電動牙刷刷洗牙本質薄片的兩側達每側1分鐘。接著以蒸餾水沖洗樣品以移除牙膏的可見痕跡,並接著儲存於人造唾液中(pH為7.4的1.5 mM CaCl2
、0.9 mM KH2
PO4
、130 mM KCl與20 mM HEPES)。可重複在人造唾液中的每個樣品的刷洗、沖洗與儲存程序每天兩次達7天,接著在37℃下乾燥樣品16小時並在進行SEM之前儲存於室溫下。
已參照各種特定實施例與技術來描述本發明。然而,在本發明的範疇內可進行許多變化例與修飾例。

Claims (10)
- 一種牙科配方,包含: 一有效量之一生物活性玻璃組成物,基於該組成物總量為100莫耳%,包含下列之一來源: 2至70%的B2 O3 , 1至60%SiO2 , 2至16%的Al2 O3 , 5至30%的CaO, 1至6%的P2 O5 , 1至20%的Na2 O, 1至20%的K2 O,以及 1至20%的MgO;以及 一有效量之一適合載體,該有效量基於該生物活性玻璃與該載體的合併重量。
- 如請求項1所述之牙科配方,其中基於該組成物總量為100莫耳%,該生物活性玻璃包含5至50%的SiO2 的一來源,且該適合載體包含1至25重量%的SiO2 的二氧化矽來源,再添加於該生物活性玻璃。
- 如請求項1或2所述之牙科配方,進一步包含:1至25莫耳%的一量的氟化物離子的一來源,再添加於該組成物,其中氟化物離子的該來源係選自NaF、SnF2 、CaF2 或前述氟化物的一組合。
- 如請求項1或2所述之牙科配方,其中該生物活性玻璃組成物的一有效量為0.1至10重量%且該適合載體的一有效量為90至99.9重量%。
- 如請求項1所述之牙科配方,其中基於該組成物總量為100莫耳%,該生物活性玻璃組成物包含: 40至60%的B2 O3 , 0.1至5%SiO2 , 6至10%的Al2 O3 , 15至25%的CaO, 1.5至2%的P2 O5 , 4至8%的Na2 O, 6至10%的K2 O,以及 6至10%的MgO。
- 如請求項5所述之牙科配方,其中該適合載體包含一或多種選自下列的成分:一研磨劑、一濕潤劑、一香料、一著色劑、一抗菌劑、一界面活性劑、一增白劑、一黏著劑或前述成分的一混合。

- 一種再礦化組成物,包含: 一有效量之一生物活性玻璃組成物,基於該組成物總量為100莫耳%,包含下列之一來源: 2至70%的B2 O3 , 1至60%SiO2 , 2至16%的Al2 O3 , 5至30%的CaO, 1至6%的P2 O5 , 1至20%的Na2 O, 1至20%的K2 O,以及 1至20%的MgO。
- 一種治療牙本質高敏性的方法,包含: 將請求項1至9中任一項所述之牙科配方與一牙齒表面接觸, 其中該牙齒表面具有下列至少一者:一牙本質表面、一牙本質小管或一牙本質表面與一牙本質的一組合。
- 如請求項9所述之方法,其中相較於不含該生物活性玻璃的一牙科配方基底,接觸該牙科配方具有50至95%的閉塞暴露的牙本質小管的一較高速率。
