Claims (18)

1. Культура клеток in vitro, содержащая: клетки человека, где по меньшей мере 10% указанных клеток человека являются незрелыми бета-клетками, экспрессирующими INS и NKX6.1: и Y-27632 в эффективном количестве, способствующем выживанию указанных незрелых бета-клеток в указанной культуре клеток.
2. Культура клеток по п. 1, отличающаяся тем, что указанная культура клеток находится в полупроницаемом инкапсулирующем устройстве.
3. Культура клеток по п. 1 или 2, отличающаяся тем, что указанные незрелые бета-клетки человека по существу не экспрессируют NGN3.
4. Криоконсервированная культура клеток in vitro, содержащая клетки человека, где указанные клетки человека содержат суспензионные агрегаты унипотентных незрелых бета-клеток человека и где по меньшей мере 10% клеток в указанных агрегатах являются незрелыми бета-клетками, экспрессирующими INS и NKX6.1; и криоконсервирующий агент.
5. Культура клеток по п. 4, отличающаяся тем, что указанные суспензионные агрегаты унипотентных незрелых бета-клеток человека находятся в полупроницаемом инкапсулирующем устройстве.
6. Культура клеток по п. 4 или 5, отличающаяся тем, что указанные суспензионные агрегаты имеют размер от 50 до 200 микрон.
7. Криоконсервированная культура клеток in vitro, содержащая: клетки человека, где по меньшей мере 10% указанных клеток человека являются незрелыми бета-клетками, экспрессирующими INS и NKX6.1; и криоконсервирующий агент.
8. Культура клеток по п. 7, отличающаяся тем, что указанные незрелые бета-клетки человека по существу не экспрессируют NGN3.
9. Культура клеток по п. 7 или 8, отличающаяся тем, что указанные незрелые бета-клетки находятся в полупроницаемом инкапсулирующем устройстве.
10. Культура клеток по любому из пп. 7-9, отличающаяся тем, что указанные незрелые бета-клетки находятся в перфорированном инкапсулирующем устройстве.
11. Культура клеток по любому из пп. 7-10, отличающаяся тем, что указанные незрелые бета-клетки человека дополнительно экспрессируют панкеротический диоденальный гомеобокс фактор - 1 (PDX-1).
12. Культура клеток по любому из пп. 7-11, отличающаяся тем, что указанные незрелые бета-клетки человека обладают способностью к созреванию в зрелые бета-клетки.
13. Культура клеток по любому из пп. 7-12, отличающаяся тем, что указанные незрелые бета-клетки дополнительно экспрессируют MAFB.
14. Культура клеток по любому из пп. 7-13, дополнительно содержащая хромогранин A (CHGA) - отрицательные клетки.
15. Культура клеток по любому из пп. 7-14, дополнительно содержащая соединение, выбранное их группы, включающей костный морфогенный белок, белок "sonic hedgehog", фактор роста тромбоцитов, член суперсемейства FGF, ретиноевую кислоту, аналоги ретиноевой кислоты, инсулиноподобный фактор роста, производное витамина В3, и никотинамид.
16. Культура клеток по любому из пп. 7-15, дополнительно содержащая матрикс.
17. Криоконсервированная культура клеток in vitro, содержащая: клетки человека, где по меньшей мере 10% указанных клеток человека являются незрелыми бета-клетками, экспрессирующими INS и NKX6.1; и криоконсервирующий агент.
18. Культура клеток по п. 17, отличающаяся тем, что указанные незрелые бета-клетки находятся в полупроницаемом инкапсулирующем устройстве.
