Claims (11)
1. Способ получения продукта ферментативного конъюгирования двух полипептидов, включающий следующие стадии:
инкубирование
i) первого полипептида, содержащего аминокислотную последовательность LPXTG (SEQ ID NO: 20, где Х может быть любым аминокислотным остатком),
ii) второго полипептида, который содержит i) аланинильное соединение на его N-конце или ii) олигоаланин Am (m равен 2 (SEQ ID NO: 26) или 3 (SEQ ID NO: 27), или 4 (SEQ ID NO: 28), или 5 (SEQ ID NO: 29)), или iii) аминокислотный остаток цистеина с последующими одним-тремя аминокислотными остатками аланина на его N-конце,
iii) третьего полипептида с сортазной активностью, который получен из сортазы А Staphylococcus aureus, и
выделение конъюгата из реакционной смеси и, посредством этого, получение продукта ферментативного конъюгирования двух полипептидов.
2. Способ по п. 1, в котором первый полипептид содержит на его С-конце аминокислотную последовательность LPXTG (SEQ ID NO: 20, где Х может быть любым аминокислотным остатком).
3. Способ по п. 2, в котором первый полипептид содержит на его С-конце аминокислотную последовательность LPETG (SEQ ID NO: 30).
4. Способ по любому из пп. 1-3, в котором второй полипептид имеет на его N-конце олигоаланин SEQ ID NO: 26 или SEQ ID NO: 27.
5. Способ по любому из пп. 1-4, в котором первый полипептид и второй полипептид независимо друг от друга выбраны из вариабельного домена антитела, фрагмента Fab тяжелой цепи антитела, области Fc антитела, метки, пептида, содержащего аминокислотную последовательность LPXTG (SEQ ID NO: 20, где Х может быть любым аминокислотным остатком), линкера и группировки несортазного мотива.
6. Способ по любому из пп. 1-5, в котором третий полипептид имеет аминокислотную последовательность SEQ ID NO: 21.
