Abstract
1. Способ получения популяции клеток-предшественников эндокринных клеток поджелудочной железы из плюрипотентных стволовых клеток, состоящий из стадий:a. Культивирования популяции плюрипотентных стволовых клеток;b. Дифференцирования популяции плюрипотентных стволовых клеток в популяцию клеток, экспрессирующих маркеры, характерные для линии сформированной эндодермы;с. Дифференцирования популяции клеток, экспрессирующих маркеры, характерные для линии сформированной эндодермы, в популяцию клеток первичной кишечной трубки;D. Дифференцирования популяции клеток первичной кишечной трубки в популяцию клеток заднего сегмента передней кишки; иE. Культивирования популяции клеток задней части передней кишки в среде с добавлением агента, расщепляющего ретиноевую кислоту.2. Способ по п. 1, в котором агент, расщепляющий ретиноевую кислоту, представляет собой ингибитор CYP26A.3. Способ по п. 2, в котором ингибитор CYP26A используют в концентрации от приблизительно 1 нмоль/л до приблизительно 1000 нмоль/л.4. Способ по п. 2, в котором ингибитор CYP26A используют в концентрации от приблизительно 10 нмоль/л до приблизительно 100 нмоль/л.5. Способ по п. 2, в котором ингибитор CYP26A представляет собой N-{4-[2-этил-1-(1H-1, 2, 4-триазол-1-ил)бутил]фенил}-1, 3-бензотиазол-2-амин.
Claims (5)
1. Способ получения популяции клеток-предшественников эндокринных клеток поджелудочной железы из плюрипотентных стволовых клеток, состоящий из стадий:
a. Культивирования популяции плюрипотентных стволовых клеток;
b. Дифференцирования популяции плюрипотентных стволовых клеток в популяцию клеток, экспрессирующих маркеры, характерные для линии сформированной эндодермы;
с. Дифференцирования популяции клеток, экспрессирующих маркеры, характерные для линии сформированной эндодермы, в популяцию клеток первичной кишечной трубки;
D. Дифференцирования популяции клеток первичной кишечной трубки в популяцию клеток заднего сегмента передней кишки; и
E. Культивирования популяции клеток задней части передней кишки в среде с добавлением агента, расщепляющего ретиноевую кислоту.
2. Способ по п. 1, в котором агент, расщепляющий ретиноевую кислоту, представляет собой ингибитор CYP26A.
3. Способ по п. 2, в котором ингибитор CYP26A используют в концентрации от приблизительно 1 нмоль/л до приблизительно 1000 нмоль/л.
4. Способ по п. 2, в котором ингибитор CYP26A используют в концентрации от приблизительно 10 нмоль/л до приблизительно 100 нмоль/л.
5. Способ по п. 2, в котором ингибитор CYP26A представляет собой N-{4-[2-этил-1-(1H-1, 2, 4-триазол-1-ил)бутил]фенил}-1, 3-бензотиазол-2-амин.
