Abstract
Description
The invention relates to the biology of tumor cells, in particular to experimental oncology.
A known method of suppressing metastasis of the tumor into the lungs, based on the use of carboxamide-quinoline LS2616. The drug is administered to mice within four days after intravenous inoculation of tumor cells of melanoma B-16-F-10. The number of pulmonary metastases is reduced by 85%
The disadvantage of this method is the side effects of vomiting, anorexia, hyperthermia and other complications due to the toxicity of the drug.
The aim of the invention is to prevent complications.
The goal is ensured by the fact that, according to the proposed method, including intravenous administration of tumor cells and counting the number of emerging pulmonary metastases, animals immediately after the introduction of tumor cells are placed in conditions of artificial dimming with illumination of 0-20 lux for 18 days before the formation of metastases. The absence of complications is associated with the safety of dark conditions compared with chemotherapy.
The experiments were performed on 139 white outbred rats, males, weighing 100-120 g. The tumor cells of rat rhabdomyosarcoma (RA-2) were inoculated with animals. The number of pulmonary metastases visible to the eye was taken into account on the 18th day after the transplantation of the tumor, the mass of which during these periods varied from 1 to 100 or more mg.
To create certain conditions of the dark regime, the windows in the vivarium's rooms were darkened with black curtains and the light intensity was measured with a luxometer.
The method is as follows.
A suspension of single tumor cells was prepared from pulmonary metastases of rat rhabdomyosarcoma (RA-2) obtained on day 18 after intravenous tumor transplantation into intact rats. For this, four animals with metastases are killed and under sterile conditions the chest is opened. The recovered lungs are transferred to Petri dishes with 199 medium or Hanks solution. To obtain a suspension of tumor cells using 30-40 metastases of different weights taken from four animals. The resulting cell suspension is injected into the tail vein of the test animals 10,000 cells in a volume of 0.2 ml of medium. After inoculation of the tumor, one group of rats was placed in a room with complete darkness (0 lux), the other group of animals was left in normal daylight. On day 18, animals are killed for counting metastases in the lungs. The number of pulmonary metastases in the experimental group decreased by 65% compared with the control group and amounted to 29.1 ± 7.4 and 84.7 ± 15, respectively. The difference is significant (P <0.01).
In contrast to the use of chemotherapeutic drugs accompanied by complications, under the influence of darkness, tumor-bearing animals did not exhibit mass loss, adynamia, seizures, hyperthermia, dyspeptic symptoms, as well as disturbances in the blood system, such as leukopenia, agranulocytosis, and thrombocytopenia.
The proposed method of suppressing tumor metastasis in comparison with known methods is the most affordable and less laborious, since it does not require any equipment, drugs or other economic costs, as well as additional manipulations. This method allows with minimal labor and material resources to obtain effective suppression of tumor metastasis to the lungs.
