A PRESENTE INVENÇÃO REFERE-SE A MONÓCITOS E MACRÓFAGOS D PARA UTILIZAÇÃO NO TRATAMENTO DA HÉRNIA DO DISCO INTERVERTEBRAL (IVD). PARA ALÉM DISSO, TAMBÉM É DIVULGADO O FENÓTIPO DE MACRÓFAGOS QUE É MAIS EFICAZ PARA UMA IMUNOTERAPIA BASEADA NA FAGOCITOSE. DESTE MODO, A PRESENTE INVENÇÃO DESCREVE COMPOSIÇÕES DE MONÓCITOS E MACRÓFAGOS, ASSIM COMO A SUA FORMA DE ADMINISTRAÇÃO. A PRESENTE INVENÇÃO DESCREVE AINDA O MÉTODO DE DIFERENCIAÇÃO, POLARIZAÇÃO E PURIFICAÇÃO PARA OBTENÇÃO DO FENÓTIPO DE MACRÓFAGOS MAIS EFICAZ. OUTRO ASPETO DESTA INVENÇÃO REFERE-SE ÀS COMPOSIÇÕES UTILIZADAS NA REFERIDA POLARIZAÇÃO DO FENÓTIPO DE MACRÓFAGO MAIS EFICAZ. AINDA OUTRO ASPETO DA PRESENTE INVENÇÃO REFERE-SE ÀS COMPOSIÇÕES UTILIZADAS NA PURIFICAÇÃO, POR SELEÇÃO NEGATIVA, DO FENÓTIPO DE MACRÓFAGO MAIS EFICAZ. OS USOS, COMPOSIÇÕES E MÉTODOS DA PRESENTE INVENÇÃO PODEM SER UTILIZADOS DE FORMA VANTAJOSA NUMA TERAPIA BASEADA EM CÉLULAS, PRINCIPALMENTE PARA USO NO TRATAMENTO DA HÉRNIA DO DISCO INTERVERTEBRAL (IVD), BEM COMO PARA OUTRAS CONDIÇÕES CLÍNICAS MELHORADAS PELA REGRESSÃO DE TECIDOS REATIVOS ADVERSOS À REGENERAÇÃO, TAIS COMO FERIDAS, TECIDO CICATRICIAL RELACIONADO COM CIRURGIAS, FIBROSE E NECROSE; A REFERIDA CONDIÇÃO CLÍNICA ESTÁ A SER SELECIONADA DA LISTA QUE INCLUÍ ENFARTE DO MIOCÁRDIO, ENDOMETRIOSE, ASMA, CIRROSE HEPÁTICA, LESÃO DA MEDULA ESPINAL E LESÕES DA CARTILAGEM.THE PRESENT INVENTION RELATES TO MONOCYTES AND MACROPHAGES D FOR USE IN THE TREATMENT OF INTERVERTEBRAL DISC HERNIATION (IVD). IN ADDITION, THE MACROPHAGE PHENOTYPE WHICH IS MOST EFFECTIVE FOR IMMUNOTHERAPY BASED ON PHAGOCYTOSIS IS ALSO DISCLOSED. THUS, THE PRESENT INVENTION DESCRIBES COMPOSITIONS OF MONOCYTES AND MACROPHAGES, AS WELL AS THEIR METHOD OF ADMINISTRATION. THE PRESENT INVENTION FURTHER DESCRIBES THE DIFFERENTIATION, POLARIZATION AND PURIFICATION METHOD FOR OBTAINING THE MOST EFFECTIVE MACROPHAGE PHENOTYPE. ANOTHER ASPECT OF THIS INVENTION REFERS TO THE COMPOSITIONS USED IN SAID POLARIZATION OF THE MOST EFFECTIVE MACROPHAGE PHENOTYPE. YET ANOTHER ASPECT OF THE PRESENT INVENTION REFERS TO COMPOSITIONS USED IN PURIFICATION, BY NEGATIVE SELECTION, OF THE MOST EFFECTIVE MACROPHAGE PHENOTYPE. THE USES, COMPOSITIONS AND METHODS OF THE PRESENT INVENTION MAY BE ADVANTAGEOUSLY USED IN CELL-BASED THERAPY, PRIMARY FOR USE IN THE TREATMENT OF INTERVERTEBRAL DISC (IVD) HERNIATION, AS WELL AS FOR OTHER CLINICAL CONDITIONS IMPROVED BY REACTIVE TISSUE REGRESSION ADVERSE TO REGENERATION , SUCH AS WOUNDS, SCAR TISSUE RELATED TO SURGERY, FIBROSIS AND NECROSIS; SAID CLINICAL CONDITION IS BEING SELECTED FROM THE LIST WHICH INCLUDES MYOCARDIAL INFARCTION, ENDOMETRIOSIS, ASTHMA, LIVER CIRRHOSIS, SPINAL CORD INJURY AND CARTILAGE INJURY.
