Abstract

Se proporciona un biosensor de actividad proteasa fiable para la determinacion de la escision de un sitio de reconocimiento de una proteasa que puede ser visualizado por FRET, cambio de fluorescencia o por electroforesis SDS-PAGE. Este biosensor consiste en un dominio globular, un conector flexible con una zona de reconocimiento/escision proteasa variable y un segundo dominio globular. La propuesta puede ser util para detectar proteasas y evaluar su actividad, como para hallar nuevos inhibidores de proteasas o monitorear la eficacia de agentes terapeuticos en pacientes en tratamiento. El biosensor propuesto es una proteina ingenearizada proveniente de la ingenierizacion racional de proteinas lo que convierte a esta molecula en unica y no presente en la naturaleza.
