Description
Claims (14)
- (a)리간드를 다공성 지지체 내로 확산시키는 단계; 및 (b) 상기 리간드를 상기 다공성 지지체에 신속하게 공유 커플링(coupling)시키기 위하여 조건을 변경하는 단계를 포함하는, 리간드를 다공성 지지체에 커플링시키는 방법에 있어서, 단계(a)중의 리간드 커플링 속도가 단계(b)중의 리간드 커플링 속도보다 낮고, 단계 (a)에서 단계(b)로 변경된 조건이 pH증가, 이온 농도 증가,또는 온도 증가, 또는 이들의 조합을 포함하는 방법.
- 제1항에 있어서, 단계(a)의 확산 조건의 pH가 단계(b)의 커플링 조건의 pH보다 낮고, 단계(a)의 확산조건의 pH가 약 3 내지 약 7의 범위이며, 단계(b)의 커플링 조건의 pH가 약 7 내지 약 10의 범위인 방법.
- 제1항에 있어서, 단계(a)의 확산 조건의 이온 농도가 단계(b)의 커플링 조건의 이온 농도 보다 낮고, 단계(a)의 이온 농도에서 단계(b)의 이온 농도로 변화하는 양이 약 0.5M 내지 약 1.5M인 방법.
- 제3항에 있어서, 단계(a)의 온도가 단계(b)의 온도보다 낮고, 단계(a)의 온도에서 단계(b)의 온도로 변환하는 정도가 약 10℃내지 약 35℃인 방법.
- 제1항에 있어서, 다공성 지지체에 리간드를 커플링 시키는 공간 분포를 조절하는 수단을 제공하고, 리간드를 다공성 지지체의 내부 표면 및 외부 표면에 공유 커플링시키는 방법을 제공하는 방법.
- 제1항 내지 제6항 중 어느 한 항에 있어서, 단계(b)의 커플링 조건이 0.1M 내지 10M 범위의 농도로 커플링 경쟁제를 사용하는 방법.
- 제1항에 있어서, 상기 다공성 지지체가 아즐락톤-작용성인 방법.
- 제1항에 있어서, 단계(a)가 단계(b)에소 보다 상대적으로 낮은 티엘 모듈러스(thiele modulus)를 갖는 방법.
- (1)지지체의 표면에 커플링된 리간드의 공간 분포를 제공하도록 제1항 내지 제8항의 방법의 단계(a) 및 (b)에 따라 다공성 지지체에 리간드를 커플링시키는 단계; 및 (2) 리게이트(ligate)를 공간적으로 분포된 리간드레 결합시키는 단계를 포함하는 다공성 지지체로의 제한된 확산 또는 다공성 지지체에 커플링된 리간드로의 결합의 입체 효과에 의해 영향받지 않도록 리게이트를 결합시키는 방법.
- 제1항 내지 제8항 중 어느 한 항의 방법에 따라 제조된 유도된 다공성 지지체.
- 커플링된 리드간의 30%이상이 다공성 지지체의 기하학적 중심의 70%이내인 내부 표면에 커플링되는 방식으로 반응성 다공성 지지체에 직접적으로 커플링되는 리간드를 포함하는 유도된 다공성 지지체.
- 80%이상의 커플링 효율로 반응성 다공성 지지체에 직접적으로 커플링되고 약 3.0결합 면역 글로블린/결합된 단백질 A 이상의 면역 글로블린에 대한 작용 효율을 갖는 단백질 A를 포함하는 유도된 다공성 지지체.
- 70%이상의 커플링 효율로 반응성 다공성 지지체에 직접적으로 커플링되고, 약 20%이상의 리게이트 단백질의 리게이트 단백질 대항체의 몰비가 2:1에서 결합하는 비율로 작용 효율을 갖는 항체를 포함하는 유도된 다공성 지지체.
- 제11항 내지 제13항 중 어느 한 항에 있어서, 상기 리간드라 아즐락톤-작용 반응성 부위를 통하여 다공성 지지체에 커플링하는 유도된 다공성 지지체※참고사항: 최초출원 내용에 의하여 공개하는 것임.
