Description
Claims (23)
- 적어도 하나의 아미노산 변형을 포함하는 조작된 클로스트리디움 독소이며, 상기 적어도 하나의 아미노산 변형이 조작된 클로스트리디움 독소의 등전점 (pI)을 상기 적어도 하나의 아미노산 변형이 없는 (그렇지 않다면) 동일한 클로스트리디움 독소의 pI보다 적어도 0.2 pI 단위만큼 더 높은 값으로 증가시키는 것인 조작된 클로스트리디움 독소.
- 제1항에 있어서, 상기 적어도 하나의 아미노산 변형이 조작된 클로스트리디움 독소의 등전점 (pI)을 상기 적어도 하나의 아미노산 변형이 없는 (그렇지 않다면) 동일한 클로스트리디움 독소의 pI보다 적어도 0.5 pI 단위만큼 더 높은 값으로 증가시키는 것인 조작된 클로스트리디움 독소.
- 제1항에 있어서, 상기 적어도 하나의 아미노산 변형이 조작된 클로스트리디움 독소의 등전점 (pI)을 상기 적어도 하나의 아미노산 변형이 없는 (그렇지 않다면) 동일한 클로스트리디움 독소의 pI보다 적어도 1 pI 단위만큼 더 높은 값으로 증가시키는 것인 조작된 클로스트리디움 독소.
- 제1항에 있어서, 상기 적어도 하나의 아미노산 변형이 조작된 클로스트리디움 독소의 등전점 (pI)을 상기 적어도 하나의 아미노산 변형이 없는 (그렇지 않다면) 동일한 클로스트리디움 독소의 pI보다 적어도 2 pI 단위만큼 더 높은 값으로 증가시키는 것인 조작된 클로스트리디움 독소.
- 제1항에 있어서, 상기 적어도 하나의 아미노산 변형이 조작된 클로스트리디움 독소의 등전점 (pI)을 상기 적어도 하나의 아미노산 변형이 없는 (그렇지 않다면) 동일한 클로스트리디움 독소의 pI보다 2 내지 5 pI 단위만큼 더 높은 값으로 증가시키는 것인 조작된 클로스트리디움 독소.
- 제1항 내지 제5항 중 어느 한 항에 있어서, 적어도 6의 pI를 갖는 조작된 클로스트리디움 독소.
- 제1항 내지 제6항 중 어느 한 항에 있어서, 6 내지 10의 pI를 갖는 조작된 클로스트리디움 독소.
- 제7항에 있어서, 7 내지 9, 바람직하게는 8 내지 9의 pI를 갖는 조작된 클로스트리디움 독소.
- 제1항 내지 제8항 중 어느 한 항에 있어서, 적어도 하나의 아미노산 변형이 아미노산 치환, 아미노산 삽입 및 아미노산 결실로부터 선택되는 것인 조작된 클로스트리디움 독소.
- 제9항에 있어서, 적어도 하나의 아미노산 치환이 산성 아미노산 잔기의 염기성 아미노산 잔기로의 치환, 산성 아미노산 잔기의 비하전된 아미노산 잔기로의 치환, 및 비하전된 아미노산 잔기의 염기성 아미노산 잔기로의 치환으로부터 선택되는 것인 조작된 클로스트리디움 독소.
- 제1항 내지 제10항 중 어느 한 항에 있어서, 1 내지 80개의 아미노산 변형을 포함하는 조작된 클로스트리디움 독소.
- 제11항에 있어서, 4 내지 40개의 아미노산 변형을 포함하는 조작된 클로스트리디움 독소.
- 제1항 내지 제12항 중 어느 한 항에 있어서, 적어도 하나의 아미노산 변형이 클로스트리디움 독소 수용체 결합 도메인 (HC 도메인)에 위치하는 것인 조작된 클로스트리디움 독소.
- 제13항에 있어서, 적어도 하나의 아미노산 변형이 클로스트리디움 독소 HCN 도메인에 위치하는 것인 조작된 클로스트리디움 독소.
- 제1항 내지 제14항 중 어느 한 항에 있어서, 적어도 하나의 아미노산 변형이 표면 노출된 아미노산 잔기의 변형인 조작된 클로스트리디움 독소.
- 제1항 내지 제15항 중 어느 한 항에 있어서, 적어도 하나의 아미노산 변형이 아스파르트산 잔기, 글루탐산 잔기, 히스티딘 잔기, 세린 잔기, 트레오닌 잔기, 아스파라긴 잔기, 글루타민 잔기, 시스테인 잔기 또는 티로신 잔기로부터 선택되는 아미노산 잔기의 변형을 포함하는 것인 조작된 클로스트리디움 독소.
- 제16항에 있어서, 아미노산 잔기가 리신 잔기 또는 아르기닌 잔기로 치환되는 것인 조작된 클로스트리디움 독소.
- 제1항 내지 제17항 중 어느 한 항에 따른 조작된 클로스트리디움 독소를 코딩하는 핵산 서열을 포함하는 핵산.
- 제18항에 따른 핵산을 적합한 숙주 세포에서 발현시키고, 숙주 세포를 용해시켜 경쇄 및 중쇄를 갖는 단일-쇄 조작된 클로스트리디움 독소 단백질을 함유하는 숙주 세포 균질물을 제공하고, 상기 단일-쇄 조작된 클로스트리디움 독소 단백질을 단리하는 것을 포함하는, 경쇄 및 중쇄를 갖는 단일-쇄 조작된 클로스트리디움 독소 단백질의 제조 방법.
- 제19항의 방법에 의해 수득가능한 단일-쇄 조작된 클로스트리디움 독소 단백질을 제공하고, 경쇄와 중쇄 사이에 위치한 인식 부위 (절단 부위)에서 폴리펩티드를 절단하는 프로테아제와 폴리펩티드를 접촉시키고, 폴리펩티드를 경쇄 및 중쇄가 디술피드 결합에 의해 함께 연결된 2쇄 폴리펩티드로 전환시키는 것을 포함하는, 조작된 클로스트리디움 독소의 활성화 방법.
