Description
도 2는 인간 MyD88을 발현시키는 HEK293XL 세포에서 본 발명에 따른 대표적인 인간 MyD88 안티센스 올리고누클레오티드의 활성을 그래프로 나타낸 것이다. 이러한 데이타는 실시예 2에 따라 배양되고 처리된 HEK293 세포에서 MyD88 발현 및 활성화를 억제하기 위한 본 발명에 따른 대표적인 올리고누클레오티드의 능력을 설명한다.

- MyD88 mRNA (SEQ ID NO: 153)에 상보적인 길이가 20 내지 50개의 누클레오티드인 합성 안티센스 올리고누클레오티드로서, 상기 안티센스 올리고누클레오티드는 SEQ ID NO: 4, 10, 21, 29, 31, 39, 46, 48, 63, 66, 70, 71, 72, 76, 85, 116 또는 142를 포함하는 서열을 가지며, 이러한 올리고누클레티드는 인간 MyD88에 특이적으로 하이브리드화하고 인간 MyD88의 발현을 억제하는, 합성 안티센스 올리고누클레오티드.
- 제 1항에 있어서, 상기 올리고누클레오티드가 알킬포스포네이트, 포스포로티오에이트, 포스포로디티오에이트 및 메틸포스포네이트로 구성된 군으로부터 선택된 하나 이상의 개질된 누클레오시드간 결합(internucleoside linkage)을 갖는 안티센스 올리고누클레오티드.

- 제 17항 또는 제 18항에 있어서, 상기 화합물이 다른 경우에는 MyD88-매개된 면역 반응을 활성화시키는 면역자극성 모티프를 포함하는 하나 이상의 비-MyD88 안티센스 올리고누클레오티드인 방법.
- 제 7항 내지 제 18항 중 어느 한 항에 있어서, 투여 경로가 비경구, 근육내, 피하, 복막내, 정맥내, 점막 전달, 경구, 설하, 경피, 국소, 흡입, 비강내, 에어로졸, 안구내, 기관내, 직장내, 질, 유전자 총(gene gun), 피부 패치, 점안약(eye drop) 또는 구강세척제(mouthwash)로부터 선택된 방법.
- 제 7항 내지 제 18항 중 어느 한 항에 있어서, 하나 이상의 백신, 항원, 항체, 세포독성제, 알레르겐, 항생제, 안티센스 올리고누클레오티드, TLR 효능제, TLR 길항제, siRNA, miRNA, 안티센스 올리고누클레오티드, 앱타머(aptamer), 단백질, 유전자 치료 벡터(gene therapy vector), DNA 백신, 애주번트, 공동 자극 분자(co-stimulatory molecule) 또는 이들의 조합물을 추가로 투여함을 포함하는 방법.
- 포유동물에 MyD88 mRNA에 상보적인 안티센스 올리고누클레오티드와 MyD88 단백질의 길항제를 투여함을 포함하여, 포유동물에서 MyD88 발현 및 활성을 억제하는 방법.
- 제 26항에 있어서, 상기 MyD88 길항제가 항-MyD88 항체 또는 이의 결합 단편 또는 펩티도미메틱(peptidomimetic), RNA-기반 화합물, 올리고누클레오티드-기반 화합물, 및 MyD88 활성의 소분자 억제제로 구성된 군으로부터 선택된 방법.
- 포유동물에 MyD88 mRNA에 상보적인 안티센스 올리고누클레오티드와 TLR 2, 4, 5, 6, 7, 8 또는 9 단백질 길항제를 투여함을 포함하여, 포유동물에서 MyD88 발현 및 활성을 억제하는 방법.
