Abstract
Description
Claims (33)
- 외부 뉴클레오타이드 서열을 포함하는 형질전환된 세균 또는 그의 자손으로, 상기 외부 뉴클레오타이드 서열이 상기 세균 또는 그의 자손을 복제하고 상기 중에서 발현되며, 선택 마커에 결합되지 않는 형질전환된 세균 또는 그의 자손.
- 제 1 항에 있어서, 외부 뉴클레오타이드 서열이 플라스미드 상에 존재하는 형질전환된 세균 또는 그의 자손.
- 제 2 항에 있어서, 플라스미드가 생존에 필요한 유전자를 암호화하고, 이때 상기 생존에 필요한 유전자가 형질전환된 세균의 세균 게놈으로부터 결실된 상기 형질전환된 세균 또는 그의 자손.
- 제 2 항에 있어서, 플라스미드가 엔도솜 이탈을 암호화하는 유전자를 함유하는 형질전환된 세균 또는 그의 자손.
- 제 4 항에 있어서, 엔도솜 이탈을 암호화하는 유전자가 pfo인 형질전환된 세균 또는 그의 자손.
- 제 2 항에 있어서, 외부 뉴클레오타이드 서열이 엔도솜 이탈을 암호화하는 형질전환된 세균 또는 그의 자손.
- 제 6 항에 있어서, 뉴클레오타이드 서열이 pfo를 암호화하는 형질전환된 세균 또는 그의 자손.
- 제 2 항에 있어서, 외부 뉴클레오타이드 서열이 항원 85a, 항원 85b, 또는 항원 85a/85b를 암호화하는 형질전환된 세균 또는 그의 자손.
- 제 2 항에 있어서, 플라스미드가 상기 플라스미드를 유지하고/하거나 안정화하는 단백질을 암호화하는 유전자를 함유하는 형질전환된 세균 또는 그의 자손.
- 제 9 항에 있어서, 단백질을 암호화하는 유전자가 항원 85a, 항원 85b 또는 항원 85a/85b를 암호화하는 형질전환된 세균 또는 그의 자손.
- 제 1 항에 있어서, 마이코박테리움인 형질전환된 세균 또는 그의 자손.
- 제 1 항에 있어서, 외부 뉴클레오타이드 서열이 세포자멸을 암호화하는 형질전환된 세균 또는 그의 자손.
- 제 2 항에 있어서, 플라스미드가 세포자멸을 암호화하는 유전자를 함유하는 형질전환된 세균 또는 그의 자손.
- 제 1 항에 있어서, 외부 뉴클레오타이드 서열이 그람 음성 세균에서 복제될 수 없는 형질전환된 세균 또는 그의 자손.
- 제 1 항에 있어서, 영양요구성인 형질전환된 세균 또는 그의 자손.
- 제 1 항에 있어서, 외부 뉴클레오타이드 서열이 일방 셔틀 벡터(one-way shuttle vector)의 적어도 일부인 형질전환된 세균 또는 그의 자손.
- 세균의 형질전환 방법으로, 상기 세균을 복제하고 상기 중에서 발현되는 외부 뉴클레오타이드 서열을 통합시키는 단계를 포함하며, 이때 상기 외부 뉴클레오타이드 서열이 선택 마커에 결합되지 않는 방법.
- 제 17 항에 있어서, 통합 단계를 일렉트로포레이션에 의해 수행하는 방법.

- 관심 유전자를 암호화하는 외부 뉴클레오타이드 서열을 포함하고, 하기의 조건들 중 하나 이상이 존재하는 형질전환된 마이코박테리움 또는 그의 자손:a) 상기 형질전환된 마이코박테리움이 그람 음성 세균에서 복제할 수 없는 플라스미드를 포함하고;b) 상기 형질전환된 마이코박테리움이 항생제 내성을 나타내지 않고;c) 상기 형질전환된 마이코박테리움이 영양요구성이고;d) 상기 형질전환된 마이코박테리움이 일방 셔틀 벡터를 함유한다.
- 제 24 항에 있어서, 외부 뉴클레오타이드 서열이 플라스미드의 일부인 형질전환된 마이코박테리움 또는 그의 자손.
- 제 25 항에 있어서, 플라스미드에 선택 가능한 마커가 결여된 형질전환된 마이코박테리움 또는 그의 자손.
- 제 25 항에 있어서, 외부 뉴클레오타이드 서열이 생존에 필요한 유전자를 암호화하고, 상기 생존에 필요한 유전자가 형질전환된 마이코박테리움의 세균 게놈으로부터 결실된 형질전환된 마이코박테리움 또는 그의 자손.
- 제 27 항에 있어서, 생존에 필요한 유전자가 leuD인 형질전환된 마이코박테리움 또는 그의 자손.
- 제 27 항에 있어서, 생체 내에서 활성화된 프로모터 서열을 추가로 포함하는 형질전환된 마이코박테리움 또는 그의 자손.
- 제 24 항에 있어서, 감독된 형질전환된 마이코박테리움 또는 그의 자손.
- 제 30 항에 있어서, BCG인 형질전환된 마이코박테리움 또는 그의 자손.
- 제 31 항에 있어서, BCG가 하기 균주 BCG1331, BCG 파스퇴르, BCG 토쿄, 및 BCG 코펜하겐 중에서 선택되는 형질전환된 마이코박테리움 또는 그의 자손.
