Claims (4)
- 전자 수용체의 존재 하에서 L-소르보손을 하기의 물리화학적 물성을 갖는 정제된 L-소르보손 탈수소 효소와 접촉시키고 반응 혼합물로부터 생성된 비타민 C를 단리시킴을 포함하는 L-소르보손으로부터 비타민 C를 생성시키는 방법:(a) 분자량: 150,000 ± 6,000 Da 또는 230,000 ± 9,000 Da (2 또는 3개의 상동성 아단위(subunit)로 구성되고 각각의 아단위는 75,000 ± 3,000 Da의 분자량을 갖는다);(b) 기질 특이성: 알데하이드 화합물에서 활성;(c) 보조인자: 피롤로퀴놀린 퀴논 및 헴(heme) c;(d) 최적 pH: L-소르보손으로부터 비타민 C를 생성시키는 경우 6.4 내지 8.2;(e) 억제제: Co2+, Cu2+, Fe2+, Ni2+, Zn2+, 모노요오도아세테이트 및 에틸렌다이아민 테트라아세트산.
- 제 1 항에 있어서,L-소르보손 탈수소 효소가 균주 글루코노박터 옥시단스(Gluconobacter oxydans) DSM 4025 (FERM BP-3812), 글루코노박터 옥시단스 DSM 4025 (FERM BP-3812)와 동일한 특성을 갖는 글루코노박터 속에 속하는 미생물 또는 이들의 돌연변이체로부터 유도되는 방법.
- 제 1 항 또는 제 2 항에 있어서,반응이 약 6.4 내지 약 9.0의 pH 범위와 약 20℃ 내지 약 60℃의 온도 범위에서 약 0.5 내지 약 48 시간 동안 수행되는 방법.
- 제 1 항 내지 제 3 항 중 어느 한 항에 있어서,반응이 약 7.0 내지 약 8.2의 pH 범위와 약 20℃ 내지 약 50℃의 온도 범위에서 약 0.5 내지 약 24 시간 동안 수행되는 방법.
