Claims (14)
- 서열번호 1의 뉴클레오타이드 서열과 95% 이상 동일한 폴리뉴클레오타이드를 포함하는, 알데히드 데하이드로게나제를 코딩하는 단리된 핵산 분자.
- (a) 서열번호 1의 뉴클레오타이드 258-2084, (b) 서열번호 1의 뉴클레오타이드 351-2084, (c) 서열번호 1의 뉴클레오타이드 258-1955, 및 (d) 서열번호 1의 뉴클레오타이드 351-1955로 이루어진 군으로부터 선택된 폴리뉴클레오타이드와 95% 이상 동일한 폴리뉴클레오타이드를 포함하는, 알데히드 데하이드로게나제를 코딩하는 단리된 핵산 분자.
- (a) 서열번호 2의 아미노산 서열을 갖는 폴리펩타이드를 코딩하는 폴리뉴클레오타이드, (b) 서열번호 2의 아미노산 32-609로 이루어진 폴리펩타이드를 코딩하는 폴리뉴클레오타이드, (c) 서열번호 2의 아미노산 1-566으로 이루어진 폴리펩타이드를 코딩하는 폴리뉴클레오타이드, 및 (d) 서열번호 2의 아미노산 32-566으로 이루어진 폴리펩타이드를 코딩하는 폴리뉴클레오타이드로 이루어진 군으로부터 선택된 폴리뉴클레오타이드를 포함하는, 알데히드 데하이드로게나제를 코딩하는 단리된 핵산 분자.
- 표준 조건 하에서 제 1 항 내지 제 3 항 중 어느 한 항의 핵산 분자의 상보적 스트랜드로 하이브리드화하는, 알데히드 데하이드로게나제 활성을 갖는 폴리펩타이드를 코딩하는 단리된 핵산 분자.
- 제 1 항 내지 제 4 항 중 어느 한 항의 핵산 분자를 포함하는 발현 벡터.
- 제 5 항에 있어서,pQE, pUC, pBluescript II, pACYC177, pACYC184, pVK100 및 RSF1010으로 이루어진 군으로부터 선택된 벡터 또는 그의 유도체로부터 선택되는 발현 벡터.
- 제 5 항의 발현 벡터로 형질전환된 재조합 미생물.
- 그의 염색체 DNA 내에 통합되어 있는 제 1 항 내지 제 4 항 중 어느 한 항의 핵산 분자를 포함하는 재조합 미생물.
- 제 7 항 또는 제 8 항에 있어서,박테리아, 효모 및 식물 세포로 이루어진 군으로부터 선택되는 재조합 미생물.
- 제 9 항에 있어서,글루코노박터(Gluconobacter), 아세토박터(Acetobacter), 슈도모나스(Pseudomonas), 클렙시엘라(Klebsiella), 아시네토박터(Acinetobacter) 및 대장균(Escherichia)으로 이루어진 군으로부터 선택되는 재조합 미생물.
- 제 10 항에 있어서,글루코노박터 옥시단스(Gluconobacter oxydans) DSM 4025인 재조합 미생물.
- L-소르보손으로부터 2-케토-L-굴론산(2-KGA) 및/또는 바이타민 C를 생산하는 방법으로서,(a) 제 7 항 또는 제 8 항의 재조합 미생물을 적절한 배양 배지에서 배양하는 단계, 및 (b) 상기 배양 배지로부터 2-KGA 및/또는 바이타민 C를 회수하고 분리하는 단계를 포함하는 방법.
- L-소르보손으로부터 2-KGA를 생산하는 방법으로서,(a) 글루코노박터 옥시단스 DSM 4025를 포함하는 미생물을 적절한 배양 배지에서 배양하되, 서열번호 2로 나타내는 알데히드 데하이드로게나제를 코딩하는 유전자가 상기 미생물에서 붕괴되는 단계, 및 (b) 상기 배양 배지로부터 2-KGA를 회수하고 분리하는 단계를 포함하는 방법.
- (a) 제 1 항 내지 제 4 항 중 어느 한 항의 핵산 분자를 포함하는 재조합 미생물을 적절한 배양 배지에서 배양하는 단계, 및 (b) 상기 배양 배지로부터 알데히드 데하이드로게나제를 회수하고 분리하는 단계를 포함하는, 알데히드 데하이드로게나제의 생산 방법.
