Claims (7)
- B형 간염 바이러스(Hepatitis B virus; 이하 'HBV'라 칭한다)의 복제 수준을 탐지하기 위한 DNA의 정량방법에 있어서,HBV를 생산하는 세포주의 배양액을 95 내지 100℃에서 10분 내지 15분 동안 가열하는 가열단계;상기 가열단계에서 얻어진 시료에 HBV에 특이적인 프라이머 및 탐침자를 넣고 실시간 PCR(real-time PCR)을 실시하는 증폭단계; 및증폭과정에서 형광신호를 85 내지 86℃에서 측정하여 반응물 내의 최초 시료량을 정량하는 정량단계;들을 포함하여 이루어짐을 특징으로 하는, 실시간 PCR에 의한 HBV DNA의 고속정량방법.
- 제 1항에서 있어서,상기 HBV에 특이적인 프라이머는 하기의 서열을 갖는 것을 특징으로 하는 상기 HBV DNA의 고속정량방법.HBV2005 : 5'-TCAGCTCTGTATCGGGAAGC-3'(SEQ ID NO 5)HBV2122 : 5'-CACCCACCCAGGTAGCTAGA-3'(SEQ ID NO 6)
- 제 1항에서 있어서,상기 HBV에 특이적인 탐침자는 하기의 서열(SEQ ID NO 3)을 갖는 상기 HBV DNA의 고속정량방법.HBV core : 5'-CCTCACCATACTGCACTCAGGCAA-3'(SEQ ID NO 3)
- 제 3항에 있어서,상기 HBV에 특이적인 탐침자는6-FAM (6-Carboxyfluorescein), TET (2',7'-dichloro-6-carboxy-4,7- dichlorofluorescein), 6-JOE (6-carboxy-4',5'-dichloro-2',7'-dimethoxyfluorescein) 또는 HEX (2',4',5',7'-tetrachloro-6-carboxy-4,7-dichlorofluorescein)로 이루어지는 그룹 중에서 선택되어지는 형광물질(emitting fluorescence)이 5' 말단에 표지되고;6-TAMRA (6-carboxytetramethylrhodamine), BHQ-1,2,3 (Black Hole Quencher), 댑실 다크 형광 억제 물질(DABCYL Dark Quencher)로 이루어지는 그룹 중에서 선택되어지는 형광억제물질(Quencher)이 3' 말단에 표지되는 것을 특징으로 하는 HBV DNA의 고속 정량 방법.
- 제 1항에 있어서,상기 실시간 PCR에서의 반응물 중 프라이머의 최종농도는 150 내지 250nM인 것을 특징으로 하는 상기 HBV DNA의 고속정량방법.
- 제 1항에 있어서,상기 실시간 PCR에서 반응물 중 탐침자의 최종농도는 200 내지 300nM인 것을 특징으로 하는 상기 HBV DNA의 고속정량방법.
- 제 1 내지 9항중 어느 한 항의 방법을 이용하여 HBV의 복제를 억제하는 약물의 역가를 측정하는 방법.
