Claims (2)
- 대장균 ATCC 21148과 대장균 ATCC 21151를 세포간 형질도입에 의해 교배시켜 대장균 HT101를 획득하는 단계;상기 대장균 HT101에 돌연변이원 NTG를 처리하여 AHV에 내성을 가지는 대장균 HT201를 선발하는 단계;상기 선발된 대장균 HT201 를 이소루이신 및 DAP가 첨가되지 않은 배지에 희석 및 도말하여 생육이 느린 대장균 변이주 MT101을 선발하는 단계; 및상기 대장균 MT101을 고농도의 아스파테이트 및 호모세린이 함유된 배지에서 생육하는 단계를 거쳐 획득함을 특징으로 하는 L-트레오닌 생산 변이주 대장균 MT201 (Escherichia coliMT201)(KCTC 18048P).
- 공시 균주 코리네박테리움 글루타미쿰 (Corynebacterium glutamicum) ATCC 31831 유래의 피루베이트 카르복실라제 유전자를 플라스미드 pTrc99A에 삽입하여 구축한 재조합 플라스미드 pPC를 상기 제 1항 기재의 아스파테이트계 대사 변이주 대장균 MT201에 도입하여 제조한 L-트레오닌 생산 재조합 대장균 MT201/pPC을 포도당 및 나트륨 사이트레이트를 혼합첨가한 배지에서 유가발효배양하여 L-트레오닌을 생산함을 특징으로 하는 L-트레오닌 발효생산방법.
