Abstract
본 발명은 혈관주사제용 프로포폴 유탁액의 제조방법에 관한 것으로, 보다 구체적으로는 고압균질기를 사용하여 40,000 내지 45,000 psi에서 균질화하는 단계를 포함하는 혈관주사용 프로포폴 유탁액의 제조방법에 관한 것이다.
본 발명에 따르면, 직경 0.1 - 0.2 ㎛의 오일입자를 만들어내어 저장안정성이 더욱 향상되고 0.45 ㎛ 이하에서도 여과되어 불순물이 용이하게 제거된 혈관 주사용 프로포폴 유탁액이 제공된다.
Description
본 발명은 혈관주사제용 프로포폴 유탁액의 제조방법에 관한 것으로, 보다 구체적으로는 고압균질기를 사용하여 균질화하는 단계를 포함하여 저장 안정성이 크게 향상되고 불순물이 적은 혈관주사용 프로포폴 유탁액의 제조방법에 관한 것이다.
프로포폴(2, 6-디이소프로필페놀)은 마취제로 일반적으로 유탁액으로 제조하여 혈관주사제로 사용되고 있다. 따라서 프로포폴 유탁액은 장기간 수상과 유상이 분리되지 않는 상태, 즉 저장안정성이 요구되고 있다.
그러나 초기에 제조된 프로포폴 유탁액은 프로포폴-오일 혼합액과 유화제-물-글리세린 혼합액을 보텍스믹서(Vortex Mixer) 또는 프로펠러믹서(Propeller Mixer)와 같은 혼합기를 사용하여 균질화하였기 때문에 유상과 수상이 쉽게 분리되어 저장안정성이 매우 불량하였다. 이를 극복하기 위하여 유화보조제로서 변형된 다당체(미국특허 제 4,997,819호) 또는 수용성 비이온 계면활성제를 사용하는 방법이 시도되었으나, 이 방법에 의한 프로포폴 유탁액 역시 단시간에서 2일정도의 저장안정성을 가지는데 불과하여 저장안정성을 크게 향상시키지는 못하였다. 최근에는 마이크로플루다이저(Microfluidizer)를 사용하여 5,000 - 23,000 psi의 고압에서 균질화함으로써, 직경 0.1㎛의 오일입자를 만들어내어 2년 이상 유상과 수상의 분리가 없이 안정하게 저장할 수 있었다고 보고된 바 있다(한국특허출원 제 93-26876 호).
본 발명자들은 상기한 기존의 프로포폴 유탁액보다 저장안정성이 더욱 향상되고 불순물이 적은 프로포폴 유탁액을 제조하기 위하여, 기존의 균질기보다 고압 상태를 형성하는 DEBEE 2000(BEE사, 이스라엘)과 같은 고압균질기를 사용하여 40,000 - 45,000psi에서 균질화한 결과 직경 0.1 - 0.2㎛의 오일입자를 만들어내어 저장안정성이 더욱 향상되었고 0.45㎛ 이하에서도 여과되어 불순물 제거가 보다 용이함을 발견하여, 본 발명을 완성하게 되었다.
본 발명의 프로포폴 유탁액의 제조방법은
a) 프로포폴-대두오일 혼합액의 제조;
b) 유화제-물-글리세린 혼합액의 제조;
c) 프로포폴-대두오일 혼합액과 유화제-물-글리세린 혼합액을 고압균질기에 투입하여 5,000 psi에서 1회 균질화;
d) c)의 균질화액을 다시 고압균질기를 사용하여 40,000 - 45,000 psi에서 2회 내지 3회 균질화; 및
e) 수산화나트륨으로 pH 7.0 - 8.5로 조절하고 여과, 충전, 밀봉 및 멸균하는 것이다.
여기에서 유화제로는 대두 인지질, 난황 인지질, 정제 포스포티딜콜린 또는 이들 중 2종 이상의 혼합물이 사용될 수 있다.
본 발명의 프로포폴 유탁액의 제조방법에서는 유화제-프로포폴-대두오일 혼합액과 물-글리세린 혼합액을 고압균질기에 투입[c)]하여 균질화함으로써 별도의 유화제-프로포폴-대두오일 혼합액과 물-글리세린 혼합액의 혼합과정이 불필요한데, 이는 기존의 방법이 양 용액의 혼합과정을 필요로 한다는 점과는 다른 것이다.
본 발명을 이하의 실시예 및 실험예에 의해 더욱 상세히 설명하나, 본 발명이 이에 의해 한정되는 것은 아니다.
[실시예 1]
정제한 난황 인지질 12.0g과 글리세린 22.5g을 물 855.5ml에 가하고 20-80℃에서 분산시킨다. 따로 프로포폴 10.0g을 고순도의 대두오일 100.0g에 가하고 20-70℃에서 혼합한다. 두 혼합액을 DEBEE 2000 균질기에 따로 따로 투입하여 5,000 psi 에서 1회 균질화하고 식힌다. 이 유탁액을 다시 DEBEE 2000 균질기를 사용하여 40,000-45,000 psi에서 2회 내지 3회 균질화한다. 이 유탁액을 수산화나트륨으로 pH 7.0-8.5로 조절하고, 여과, 충전, 밀봉 및 멸균한다. 이렇게 만들어진 유탁액의 오일입자는 0.1-0.2㎛이다.
Claims (1)
- a) 프로포폴-대두오일 혼합액의 제조;b) 유화제-물-글리세린 혼합액의 제조;c) 프로포폴-대두오일 혼합액과 유화제-물-글리세린 혼합액을 고압균질기에 투입하여 5,000 psi에서 1회 균질화;d) c)의 균질화액을 다시 고압균질기를 사용하여 40,000 - 45,000 psi에서 2회 내지 3회 균질화; 및e) 수산화나트륨으로 pH 7.0 - 8.5로 조절하고 여과, 충전, 밀봉 및 멸균하는 것을 특징으로 하는 프로포폴 유탁액의 제조방법.
