Claims (103)

여기서 상기 제 2 폴리뉴클레오티드가 상기 트랜스포손 말단 서열에 상보적인 영역을 포함하며, 트랜스포솜 복합체가 동종이합체(homodimer)이고, 동종이합체가 제 1 서열의 제 1 태그를 포함하는 제 1 복수의 동종이합체 및 제 2 서열의 제 1 태그를 포함하는 제 2 복수의 동종이합체를 포함함; 및

선택적으로 상기 고정된 제 1 폴리뉴클레오티드 및 상기 용액상 제 1 폴리뉴클레오티드가 서로 다른 트랜스포손 말단 서열을 포함하고, 상기 루프형 올리고뉴클레오티드가 상기 서로 다른 트랜스포손 말단 서열의 각각에 상보적인 서열을 상기 제 1 및 제 2 말단에 포함하는 방법. - 제 16항에 있어서, 상기 고정시키는 것이 하기를 포함하는 방법:

트랜스포솜 복합체가 동종이합체이며, 동종이합체가 제 1 서열의 제 1 폴리뉴클레오티드를 포함하는 제 1 복수의 동종이합체 및 제 2 서열의 제 1 폴리뉴클레오티드를 포함하는 제 2 복수의 동종이합체를 포함하는, 미세입자의 집단. - 제 27항에 있어서, 상기 제 2 폴리뉴클레오티드가 이의 5' 말단에서 포스포릴화되는, 미세입자의 집단.
- 제 27항에 있어서, 각각의 미세입자가 복수의 상기 트랜스포솜 복합체를 포함하는, 미세입자의 집단.
- 제 29항에 있어서, 각각의 미세입자가 제 1 폴리뉴클레오티드의 제 1 및 제 2 부분집단을 포함하고, 상기 제 1 부분집단이 제 1 태그를 포함하고 상기 제 2 부분집단이 제 2 태그를 포함하는, 미세입자의 집단.
- 제 27항에 있어서, 제 1 폴리뉴클레오티드의 5' 부분이 바코드 인덱스 태그(barcode index tag)를 추가로 포함하고, 개별적인 미세입자 상의 제 1 폴리뉴클레오티드가 동일한 인덱스 태그를 공유하는, 미세입자의 집단.
- 제 31항에 있어서, 상기 집단이 상기 제 1 폴리뉴클레오티드 상에 복수의 인덱스 태그를 포함하는, 미세입자의 집단.
- 제 27항에 있어서, 상기 트랜스포솜 복합체가 과활동성 Tn5 트랜스포사제를 포함하는, 미세입자의 집단.
- 제 27항에 있어서, 상기 미세입자가 비드 또는 상자성 비드인, 미세입자의 집단.
- 하기를 포함하는, 복수의 부분-단편(sub-fragment)을 포함하는 서열 판독으로의 인덱스-지정 어셈블리를 위해 태깅된 DNA 단편의 라이브러리를 생성하는 방법:

(i) 트랜스포손 말단 서열을 포함하는 3' 부분, 및
(ii) 상기 인덱스 태그를 포함하는 5' 부분을 포함하며;
상기 제 2 폴리뉴클레오티드가 상기 트랜스포손 말단 서열에 상보적인 영역을 포함하고,
트랜스포솜 복합체가 동종이합체이며, 동종이합체가 제 1 서열의 제 1 폴리뉴클레오티드를 포함하는 제 1 복수의 동종이합체 및 제 2 서열의 제 1 폴리뉴클레오티드를 포함하는 제 2 복수의 동종이합체를 포함하고,
상기 미세입자의 집단이 적어도 복수의 인덱스 태그를 포함하며, 개별적인 미세입자 상의 제 1 폴리뉴클레오티드가 동일한 인덱스 태그를 공유함; 및
(b) 표적 DNA가 트랜스포솜 복합체에 의해 단편화되고, 제 1 폴리뉴클레오티드의 3' 트랜스포손 말단 서열이 단편의 적어도 한 가닥의 5' 말단으로 전이되는 조건 하에 표적 이중-가닥 DNA를 고체 지지체에 적용시켜; 이에 의해 이중 가닥 단편의 고정된 라이브러리를 생성하는 단계, 여기서 적어도 한 가닥이 상기 인덱스 태그로 5'- 태깅됨. - 제 35항에 있어서, 상기 미세입자로부터 상기 태깅된 DNA 단편을 유리시키는 것을 추가로 포함하는 방법.

- 하기를 포함하는, 복수의 표적 DNA 분자를 서열화하는 방법:
a) 표적 DNA가 트랜스포솜 복합체에 의해 단편화되고, 제 1 폴리뉴클레오티드의 3' 트랜스포손 말단 서열이 단편의 적어도 한 가닥의 5' 말단으로 전이되는 조건 하에 복수의 표적 이중-가닥 DNA를, 그 위에 고정된 트랜스포솜 복합체를 갖는 고체 지지체에 적용시켜; 이에 의해 이중 가닥 단편의 고정된 라이브러리를 생성하는 단계, 여기서 적어도 한 가닥이 제 1 태그로 5'-태깅되고,
상기 트랜스포솜 복합체 각각은 제 1 및 제 2 폴리뉴클레오티드를 포함하는 이중-가닥 핵산의 제 1 폴리뉴클레오티드에 비-공유적으로 결합된 트랜스포사제를 포함하며, 상기 제 1 폴리뉴클레오티드가 상기 고체 지지체에 고정되고, 상기 제 1 폴리뉴클레오티드가
(i) 트랜스포손 말단 서열을 포함하는 3' 부분, 및
(ii) 제 1 태그를 포함하는 5' 부분을 포함하며,
상기 제 2 폴리뉴클레오티드는 상기 트랜스포손 말단 서열에 상보적인 영역을 포함하고,
트랜스포솜 복합체가 동종이합체이며, 동종이합체가 제 1 서열의 제 1 폴리뉴클레오티드를 포함하는 제 1 복수의 동종이합체 및 제 2 서열의 제 1 폴리뉴클레오티드를 포함하는 제 2 복수의 동종이합체를 포함하고,
각 표적 DNA의 제 1 부분은 상기 고체 지지체의 제 1 위치에서 상기 고체 지지체에 부착되고 각 표적 DNA의 제 2 부분은 상기 고체 지지체의 제 2 위치에서 상기 고체 지지체에 부착됨; 및
b) 상기 고정된 이중 가닥 단편의 라이브러리를 맵핑시켜 각 표적 DNA에 의해 연결되는 위치의 세트를 생성하는 단계;
c) 표적 DNA의 상기 제 1 및 제 2 부분의 서열을 결정하는 단계;
d) 상기 위치의 세트를 상호관련시켜 어느 제 1 및 제 2 부분이 상기 표적 DNA에 의해 연결되는지를 결정하고 표적 DNA 분자의 서열을 결정하는 단계. - 제 47항에 있어서, 단계 c)가 상기 고정된 이중 가닥 단편을 절단하여 유리 말단(free end)을 유리시키는 것을 포함하는 방법.
- 제 48항에 있어서, 상기 절단이 절단가능한 부위를 절단하는 것을 포함하고, 선택적으로 상기 절단가능한 부위가 변형된 뉴클레오티드를 포함하거나 제한 효소 부위인 방법.
- 제 47항에 있어서, 맵핑이 고정된 DNA의 물리적 맵을 생성하는 것을 포함하는 방법.
- 제 50항에 있어서, 물리적 맵이 DNA의 영상화에 의해 생성되어, 상기 고체 표면을 가로질러 고정된 DNA 분자의 위치를 확립하는 방법.
- 제 51항에 있어서, 고정된 폴리뉴클레오티드가 영상 제제를 고체 지지체에 첨가하고 영상 제제로부터 신호를 검출함에 의해 영상화되는 방법.
