Abstract
본 발명은 레몬밤 추출물을 유효 성분으로 포함하는 지방 분해용 조성물, 그 조성물을 이용한 지방 분해용 주사제 조성물, 그 조성물을 이용한 지방 분해용 화장료 조성물에 관한 것이다.

본 발명은 레몬밤 추출물을 유효성분으로 포함하는 지방 분해용 주사제 조성물에 관한 것이다.
일반적으로, 갸름한 V라인형 얼굴은 누구나 원하는 얼굴형이다. 특히, 근래에는, 통통한 얼굴에서 벗어나 작고 윤곽이 뚜렷한 얼굴 라인을 형성하고자 하는 사람들이 많은 추세이다.
지방성형(lipoplasty)을 목적으로 포스파티딜콜린(Phosphatidylcholine, PPC라 함)과 데옥시콜린산(sodium deoxycholate)을 이용한 복합제제 주사제가 국소비만에 많이 이용되어 왔다.
하지만, 이러한 포스파티딜콜린과 데옥시콜린산을 이용한 복합제제 주사제에 의한 지방괴사 덩어리 발생으로 인해, 포스파티딜콜린을 제외한 데옥시콜린산 단독 제제를 이용한 지방분해 조성물을 개발하고 있으나, 데옥시콜린산 단독 제제를 이용한 지방분해 조성물의 경우 부종, 홍반, 통증, 피부경결 등의 부작용이 빈번하게 발생하는 문제점이 있다.
본 발명은 전술한 종래 기술의 문제점을 해결하기 위한 것으로서, 주사만으로 용이하게 얼굴이 갸름해지게 하는 주사제 조성물을 제공하는 것을 목적으로 한다.
본 발명은, 레몬밤잎 추출물을 유효성분으로 하여 도포되는 영역의 지방을 분해하는 화장료 조성물을 제공하는 것을 목적으로 한다.
본 발명의 실시예에 따르면, 레몬밤잎 추출물을 유효성분으로 하는 지방 분해용 주사제 조성물을 제공한다.
본 발명의 다른 실시예에 따르면, 레몬밤잎 추출물, 마로니에(Aesculus Hippocastanum) 추출물, 푸쿠스 베시쿨로수스(FucusVesiculosus) 추출물, 호두(JuglansRegia) 추출물, ADP(Disodium Adenosine Phosphate), Adenosine Triphosphate, Tyrosine을 포함하는 지방 분해용 조성물을 제공한다.
본 발명의 다른 실시예에 따르면, 상기 조성물을 유효성분으로 함유하는 지방 분해용 주사제 조성물을 제공한다.
본 발명의 다른 실시예에 따르면, 상기 조성물을 유효성분으로 함유하는 지방 분해용 화장료 조성물을 제공한다.
본 발명의 다른 실시예에 따르면, 소듐클로라이드 0.898중량%, 프로필렌글라이콜 0.11462중량%, 페녹시에탄올 0.0468중량%, 메틸프로판다이올 0.04중량%, 레몬밤잎 추출물 0.01875중량%, 타이로신 0.00800중량%, 가시칠엽수씨추출물 0.00560중량%, 호두추출물 0.00560중량%, 에틸헥실글리세린 0.00520중량%, 블래더랙추출물 0.00240중량%, 메틸실란올만누로네이트 0.00180중량%, 소르빅애씨드 0.00100중량%, 아데노신포스페이트 0.00024중량%, 다이소듐아데노신트라이포스페이트 0.00024중량%, 및 잔량의 정제수를 포함하는 지방 분해용 주사제 조성물을 제공한다.
본 발명에 따르면, 주사만으로 용이하게 얼굴이 갸름해지게 하는 주사제 조성물을 제공할 수 있다.
본 발명에 따르면, 레몬밤잎 추출물을 유효성분으로 하여 도포되는 영역의 지방을 분해하는 화장료 조성물을 제공할 수 있다.

이하, 본 발명의 구체적인 실시예를 첨부한 도면을 참조하여 상세히 설명한다.
본 발명의 실시예에 따른 레몬밤 추출물을 유효성분으로 포함하는 지방분해용 조성물은 지방 분해, 림프 순환, V라인 얼굴, 탄력있는 얼굴을 위해 주사제로 피하에 투약되거나 화장료 조성물로 도포될 수 있다.
본 발명의 실시예에 따른 조성물은 눈두덩 지방, 눈밑 지방, 볼살, 광대, 입가지방, 턱라인, 이중턱 등의 부위에 주사하여 지방 분해를 통해 얼굴 라인을 바로잡는 데 시술될 수 있다. 개인의 얼굴 상태에 따라 섬세한 조각이 가능한 효과적인 조성물이다. 부종완화, 지방 분해, 림프 순환 촉진, 안티 셀룰라이트 작용을 한다.
본 발명의 실시예에 따른 지방 분해 조성물은 레몬밤잎 추출물, 마로니에(Aesculus Hippocastanum) 추출물, 푸쿠스 베시쿨로수스(FucusVesiculosus) 추출물, 호두(JuglansRegia) 추출물, ADP(Disodium Adenosine Phosphate), Adenosine Triphosphate, Tyrosine을 포함하는 것이 바람직하다.
본 발명의 실시예에 따른 지방 분해 주사제 조성물은 다음 성분으로 구성되는 것이 바람직하다.

레몬밤(Melissa officinalis)은 꿀풀과(Labiatae)에 속하는 식물로, 다양한 활성 플라보노이드(시나로사이드(cynaroside), 코스모신(cosmosin), 람노시트린(rhamnocitrin), 이소퀘르시트린(isoquercitrin)), 테르펜및 트리테르펜산(triterpene acids: 우르솔산(ursolic acid) 및 올레아놀산(oleanolic acid))을 함유하고 있는 것으로 알려져 있고(Herodez SS, Hadolin M, Skerget M, Knez Z Solvent extraction study of antioxidants from balm (Melissa officinalis L) leaves Food Chemistry 200;80:275-82), 정유, 카페인산 및 로즈마린산, 모노테르펜과 세스퀴테르펜, 페놀계 물질 및 탄닌 등의 활성성분 역시 레몬밤에서 분리 추출되었다. 현재까지 레몬밤 추출물의 항패혈증(Ulbricht C, Brendler T, Gruenwald J, Kligler B, Keifer D, Abrams TR, Woods J, Boon H, Kirkwood CD, Hackman DA, Basch E, Lafferty HJ; Natural Standard Research Collaboration Lemon balm (Melissa officinalis L): an evidence-based systematic review by the Natural Standard Research Collaboration J Herb Pharmacother 2005;5:71-114), 항바이러스), 항균 및 항산화 활성이 보고되어 왔다(Bolkent S, Yanardag R, Karabulut-Bulan O, Yesilyaprak B Protective role of Melissa officinalis L extract on liver of hyperlipidemic rats: a morphological and biochemical study J Ethnopharmacol 2005;99:391-8, Ali Zarei1 , Saeed Changizi Ashtiyani, Soheila Taheri, Fateme Rasekh Comparison between effects of different doses ofofficinalisatorvastatin on the activity of liver enzymes in hypercholesterolemia rats AJP 2014; 4(1) 15-23). 레몬밤잎 추출물은 Melissa officinalis leaf extract로 지중해 연안 및 중부에 자생하며 "멜리사"라고도 불리는 허브식물로 레몬과 비슷한 향이 특징이며 풍부한 로즈마린산 성분을 함유한다. 로즈마린산은 다이어트, 항산화, 항염 작용에 도움을 준다. 레몬밤잎 추출물은 폴리페놀과 타닌 성분으로 피부 보호에도 탁월한 효능을 가진다. 레몬밤 추출물은 물, 메탄올, 에탄올 및 이들 중 2종 이상의 혼합물로 이루어지는 군으로부터 선택되는 극성용매, 바람직하게는 물로 레몬밤 전초, 바람직하게는 레몬밤의 잎을 추출하고 분무건조시켜 분말화한 것이다.
가시칠엽수씨추출물(AESCULUS HIPPOCASTANUM (HORSE CHESTNUT) SEED EXTRAC)은 항부종작용, 항염증작용, 혈관 조직 벽에 대해 부종형성방지, 급약성의 감소 뿐만 아니라 피부를 자외선으로부터 지켜 기미를 예방해주며 거친 피부를 방지하는 효과가 있다.
호두추출물(JUGLANS REGIA (WALNUT) SEED EXTRACT)은 세포에서 각질형성세포에 의한 각질층의 분화를 촉진하고, 손상된 피부의 장벽기능을 회복시키며, 보습력 증가 및 유지에 뛰어난 효과가 있을 뿐만 아니라, 비만세포에서의 탈 과립저해 및 면역세포증식 효과가 뛰어나며 항염, 자극완화 효과가 있다.
블래더랙추출물(FUCUS VESICULOSUS EXTRACT)은 큰 갈조류 해초로부터 추출한 성분으로 다량의 미네랄, 비타민, 아미노산을 함유하고 있어 피부를 보호하고 수화하며 거칠고 건조한 피부를 촉촉하게 하는 효과가 있다. 지방을 흡수하고 노폐물 배출에 도움을 준다.
소듐클로라이드(SODIUM CHLORIDE)는 소듐(나트륨)과 클로라이드(염화물)의 화합물인 무기염 성분으로 화장품 및 미용제품에서 산도 조절제, 방부기능, 향미제, 점도증진제로써 사용된다.
프로필렌글라이콜(PROPYLENE GLYCOL)은 프로필렌옥사이드를 가수분해시켜 얻어지는 성분으로 분자량이 작아 피부침투력이 우수하여 유효성분을 침투시키기 위한 용도로 사용된다. 피부 속 수분을 유지시켜 피부 보습력 향상시키는 효과가 있다.
페녹시에탄올(PHENOXYETHANOL)은 페놀과 에틸렌글라이콜이 결합한 성분으로 주로 화장품에 방부제와 보류제의 역할을 하는 성분으로 한때 유방암으로 악명을 떨쳤던 파라벤을 대신해 화장품에 사용되는 대체 방부제 중 하나이다.
메틸프로판다이올(METHYLPROPANEDIOL)은 용매로 사용되는 글라이콜의 일종이다. 각종 성분들(살리실릭산등)이 피부에 침투하도록 도움을 준다. 수화(Hydrating) 속성이 있으며, 피부를 부드럽고 촉촉하게 하는데 도움을 준다.
타이로신(TYROSINE)은 멜라닌의 생성을 시작하는 피부 아미노산이다. 체내 카테콜아민(Catecholamine), 갑상선 호르몬인 티록신(Thyroxine)의 전구체로, 지방대사를 촉진한다.
에틸헥실글리세린(ETHYLHEXYLGLYCERIN)은 글리세릴에테르로 유기화합물성분이며 글리세린의 한 종류로 부드러움, 습윤제 및 적정한 번짐 성능을 가진 습윤제의 일종이다. 페녹시에탄올, 이소티아졸리논, 그리고 파라벤 등과 같은 방부제의 항균력을 증가시켜 주어 방부제 사용량을 줄여줄 수 있다. 박테리아 성장을 억제하여 탈취효과가 있으며 피부 컨디셔닝을 부여한다.
메틸실란올만누로네이트(METHYLSILANOL MANNURONATE)은 유기 실리콘 Organic silicon(SI)인 실라놀(Silanol)과 갈조류의 알긴산에서 유래한 마누론산(Mannuronic Acid)가 결합된 물질로, 피부재생, 탄력강화, 튼살 방지 효과 및 항산화, 지방분해, 보습 효과 등이 있다.
소르빅애씨드(SORBIC ACID)는 블루베리 같은 베리류에 존재하는 성분으로 세균, 곰팡이 등 미생물의 생육 억제 효과가 있어 모든 화장품에 방부 성분으로 주로 사용 사용된다. 화장품 뿐만 아니라 식품에도 허용되는 성분이다.
아데노신포스페이트(ADENOSINE PHOSPHATE)는 복합성, 주름개선, 항염제, 진정제, 세포대화성분으로 진피층 내 피부 세포 활성화 산소를 전달하며 섬유아세포의 크기 증대 및 증식을 강화하여 콜라겐 합성을 촉진한다.
다이소듐아데노신트라이포스페이트(DISODIUM ADENOSINE TRIPHOSPHATE)는 아데노신 트리포스페이트의 디소듐염 성분이다. 피부에 컨디셔닝을 부여하는 역할로 사용된다.
광대가 발달되어 인상이 강해 보이는 경우, 나이가 들면서 심술보 턱선 등이 쳐진 경우, 팔자 주름 옆의 볼 때문에 팔자 주름이 깊어 보이는 경우, 볼이나 이중턱과 같이 부분적으로 지방이 많은 경우, 보톡스만으로는 사각턱 교정이 안될 경우, 윤곽이나 양악 수술을 했음에도 얼굴라인이 교정이 안되는 경우, 지방 흡입이나 수술을 두려워하는 경우, 지방 이식이 과한 경우, 리프팅 전/후, 안면비대칭환자, 기존 지방 분해 약물로 효과를 보지 못하는 경우, 짧은 시간에 일상생활에 지장을 주지 않으면서 갸름한 얼굴을 원하는 환자를 대상으로 한다.
주사제 투약 임상 결과는 다음과 같다.
환자 연령은 20대 이상의 남녀 환자를 대상으로 한다. 매주 1회씩 4회 한 달간 시술한 후 결과이다. 시술 방법은 30~40분 연고 마취 후, 32G needle(1cc syringe), Mesogun을 사용하였다. 레몬밤 조성물 Solution은 1cc당 3-4 point로 나눠서 피하(SQ level)에 주사 한다. 시술 후 부드럽게 마사지하고, 항생제 연고 또는 Vit. K 연고를 도포한다.
다음 표 2는 시술 부위별 시술 횟수, 용량 및 간격을 나타낸 표이다.
시술자는 80~90% 이상 효과를 보였다. 전체적인 모습중 심술보와 턱선이 가장 드라마틱하게 변화되는 모습을 보였으며 효과는 2~3주부터 확실히 나타났다. 약물사용 후 부종이나 작열감이 없었고 일상 생활을 하는데 불편함이 없었다. 실리프팅과 보톡스시술이 필요한 환자의 경우 병행 시술로 만족도가 더 높아졌다. 특히 윤곽 약물은 림프대사에도 관여하여 부종완화와 셀룰라이트 분해에 탁월한 효과를 보였다. 즉, 도 1에 도시된 바와 같이 19%가 단단한 셀룰라이트 감소를 보였고, 32%가 부종성 셀룰라이트 감소를 보였고,49%가 연한 셀룰라이트 감소 효과를 보였다.
도 2 및 3은 본 발명의 실시예에 따른 조성물이 지방 세포에 반응한 실험을 나타낸 전자 현미경 사진을 나타낸다. 도 2는 조성물 투입전 사진이고, 도 3은 조성물 투입후 사진이다. 약 80%의 지방 세포가 감소했음을 확인할 수 있다.
도 4 및 5는 본 발명의 실시예에 따른 지방 분해 조성물을 이용한 시술을 4주간 수행하기 전과 후의 사진을 각각 나타낸다. 도시된 바와 같이, 4주만에 지방 및 셀룰라이트의 분해로 드라마틱한 얼굴라인의 변화를 확인할 수 있다.
