Abstract
Description
도 2는 본 발명에 적용되는 화합물의 고리매개 등온증폭법(LAMP)에 의한 MRSA의 존재에 따른 발색 평가 결과를 나타낸 것이다. 도면에서 P는 positive control, N은 negative control을 나타낸다.
도 3은 MRSA DNA의 증폭을 확인하기 위하여 전기영동법을 실시하여 확인한 결과를 나타낸 것이다. 도면에서 P는 positive control, N은 negative control을 나타낸다.
도 4는 본 발명의 화합물의 고리매개 등온증폭법(LAMP)에 의한 MRSA DNA에 대한 민감도 평가결과를 나타낸 것이다. 도면에서 NC는 negative control을 나타낸다.
도 5는 화학식 7의 화합물의 MRSA DNA에 대한 발색 평가결과를 나타낸 것이다. 도면에서 NC는 negative control을 나타낸다.
도 6은 본 발명의 방법에 의하여 MRSA를 검출하는 것과 종래의 기술인 PCR 반응을 이용하여 MRSA를 검출하는 것의 민감도를 확인한 결과를 나타낸 것이다.

Claims (5)
- 마그네슘 이온의 농도 변화를 통하여 메티실린 내성 황색포도상구균(MRSA)의 감염을 진단할 수 있는 키트에 있어서, 상기 키트는 하기의 화학식 1로 표현되는 염료 화합물을 포함하여 이루어지는 것을 특징으로 하는 키트

상기 화학식 1에서
R1은 히드록시기이며, 상기 R2는 수소 또는 아미노이고, 상기 R2가 히드록시면 상기 R1은 아미노이며; R3은 수소이고;
R4는 수소 또는 -NHRa 중에서 선택되고, 상기 Ra는 수소, 탄소 1개 내지 6개의 알킬 및 페닐 중에서 선택되며;
R5 및 R6 중 어느 하나는 설폰산기이고 다른 하나는 수소이고;
Ar은 페닐 또는 나프틸이며, 상기 페닐 또는 나프틸의 임의의 수소 2 내지 3개는 히드록시, 니트로, 설폰산기, 할로겐 중에서 선택되는 치환기로 치환되고, 상기 임의의 수소 2 내지 3개에 치환된 치환기의 하나는 2번 탄소위치에 치환된 히드록시이며; 상기 설폰산기는 설폰산(-SO3H) 또는 설폰산나트륨염(-SO3Na)이다.
- 제1항 내지 제3항 중 선택된 어느 한 항에 있어서 상기 메티실린 내성 황색포도상구균(MRSA)은 고리매개 등온증폭법(LAMP)에 의하여 증폭이 이루어지는 것을 특징으로 하는 키트
- 하기의 화학식 2 내지 4, 7 내지 9로 표현되는 화합물 중 하나 이상을 이용하여 고리매개 등온증폭법(LAMP)을 통해 메티실린 내성 황색포도상구균(MRSA)의 감염을 검출하기 위한 방법에 있어서, 상기 검출방법은 화학식 2 내지 4, 7 내지 9로 표현되는 각각의 화합물이 LAMP 반응액 내의 마그네슘과 복합체를 형성하고, 고리매개 등온증폭(LAMP) 반응 과정에서 발생하는 마그네슘 농도 변화에 의하여 상기 화학식 2 내지 4, 7 내지 9로 표현되는 염료 화합물의 발색 파장이 서로 상이하게 변화하는 것을 특징으로 하는 육안으로 마그네슘 이온의 농도 변화를 통해 MRSA를 검출하는 방법
