Claims (47)
- a) 라이신 또는 시스테인 잔기를 포함하지 않는, 서열번호 1, 서열번호 3, 및 서열번호 5 내지 서열번호 7의 모듈로 이루어진 군으로부터 독립적으로 선택된 5 내지 50개의 모듈로 이루어진 거미 실크 단백질을 코딩하는 핵산 서열을 제공하는 단계;b) 단계 a)의 거미 실크 단백질의 1개 모듈 내의 하나의 세린 및/또는 글루타메이트를 코딩하는 핵산 서열을 하나의 라이신 및/또는 시스테인을 코딩하는 핵산 서열로 교체하여, 변형된 핵산 서열을 얻는 단계, 및선택적으로 라이신 또는 시스테인을 포함하는 서열번호 20 내지 서열번호 24의 아미노 말단 TAG를 코딩하는 핵산 서열을 상기 변형된 핵산 서열에 첨가하거나, 라이신 또는 시스테인을 포함하는 서열번호 25 내지 서열번호 28의 카르복시 말단 TAG를 코딩하는 핵산 서열을 상기 변형된 핵산 서열에 첨가하는 단계;c) 단계 b)에서 얻은 변형된 핵산 서열을 적합한 숙주 내에서 발현하는 단계; 및d) 상기 발현된 변형된 거미 실크 단백질을 회수하는 단계;를 포함하는, 거미 실크 단백질을 변형시키는 방법.
- a) 라이신 또는 시스테인 잔기를 포함하지 않는, 서열번호 1 내지 서열번호 7의 모듈로 이루어진 군으로부터 독립적으로 선택된 5 내지 50개의 모듈로 이루어진 거미 실크 단백질을 코딩하는 핵산 서열을 제공하는 단계;b) 라이신 또는 시스테인을 포함하는 서열번호 20 내지 서열번호 24의 아미노 말단 TAG를 코딩하는 핵산 서열을 상기 단계 a)에서 제공된 핵산 서열에 첨가하거나, 라이신 또는 시스테인을 포함하는 서열번호 25 내지 서열번호 28의 카르복시 말단 TAG를 코딩하는 핵산 서열을 상기 단계 a)에서 제공된 핵산 서열에 첨가하는 단계;c) 단계 b)에서 얻은 변형된 핵산 서열을 적합한 숙주 내에서 발현하는 단계; 및d) 상기 발현된 변형된 거미 실크 단백질을 회수하는 단계;를 포함하는, 거미 실크 단백질을 변형시키는 방법.
- 제1항 또는 제2항에 있어서, 상기 변형된 거미 실크 단백질 내의 상기 라이신 및/또는 시스테인 분자에 다른 물질을 결합시키는 것을 추가로 포함하는 방법.
- 제1항 또는 제2항에 있어서, 상기 라이신 및/또는 시스테인에 결합되는 다른 물질은 폴리펩티드, 폴리사카라이드, 마커 분자, 양자점(quantum dots), 금속, 핵산, 지질(lipids) 및 저분자 약물로 이루어진 군으로부터 선택되는 방법.

- 제43항에 있어서, 상기 단백질은 C16CC 또는 CCC16이고, 상기 모듈 C는 서열번호 3의 아미노산 서열이고, 모듈 CC는 서열번호 10의 아미노산 서열인 변형된 거미 실크 단백질.
