Claims (12)
- HIV NC 단백질을 암호화하며, 포유동물 세포에서 고발현되도록 코돈-최적화시킨 서열번호 5, 서열번호 8, 서열번호 10 및 서열번호 11로 표시되는 염기서열로 이루어진 군에서 선택된 HIV NC(Optimized HIV NC) 폴리뉴클레오티드.
- 삭제
- 제1항의 코돈-최적화시킨 HIV NC(Optimized HIV NC) 폴리뉴클레오티드를 포함하는 HIV NC 단백질 발현용 벡터.
- 제3항에 있어서, 상기 벡터가 a) SV40 19s mRNA 인트론 서열, 서열번호 2로 표시되는 변형된 SV40 16S mRNA 인트론 서열 및 β-글로빈 인트론 서열로 이루어진 군에서 선택된 어느 하나의 서열; 및 b) HIV NC 단백질을 암호화하며, 포유동물 세포에서 고발현되도록 코돈-최적화시킨 서열번호 5, 서열번호 8, 서열번호 10 및 서열번호 11로 표시되는 염기서열로 이루어진 군에서 선택된 HIV NC(Optimized HIV NC) 폴리뉴클레오티드가 순차적으로 연결된 것을 포함하는 것을 특징으로 하는 HIV NC 단백질 발현용 벡터.
- 제4항에 있어서, 상기 코돈-최적화시킨 HIV NC(Optimized HIV NC) 폴리뉴클레오티드 하부에 mRNA 안정화 인자(mRNA stability element)가 추가적으로 연결된 것을 특징으로 하는 벡터.
- 제4항에 있어서, 상기 SV40 19s mRNA 인트론 서열은 서열번호 1로 이루어지며, β-글로빈 인트론(β-globin intron) 서열은 서열번호 9로 이루어진 것을 특징으로 하는 벡터.
- 제4항에 있어서, 상기 HIV NC 단백질 발현용 벡터는 도 3에 개시된 pCMV(-HA)/OptiNC 벡터의 개열지도를 갖는 것을 특징으로 하는 벡터.
- 제5항에 있어서, 상기 mRNA 안정화 인자는 RRE(Rev response element), WPRE(woodchuck post-transcriptional regulatory element), 베타-액틴 3'-UTR(β-actin 3'-untranslated regions) 및 RSV 안정화 인자 (Rous Sarcoma Virus stability element)로 이루어진 군에서 선택된 어느 하나인 것을 특징으로 하는 벡터.
- 제5항에 있어서, 상기 HIV NC 단백질 발현용 벡터는 도 4에 개시된 pCMV(-HA)/OptiNC/RRE 벡터의 개열지도를 갖는 것을 특징으로 하는 벡터.
- 제3항 내지 제9항 중 어느 한 항의 벡터로 형질전환된 세포.
- 제10항에 있어서, 상기 세포가 COS-7 또는 293T 세포인 것을 특징으로 하는 형질전환된 세포.
- 제10항의 형질전환된 세포를 배양하는 단계를 포함하는 HIV NC 단백질의 생산방법.
