Abstract
Description
포피 지지세포층-인간 ES세포는 미국특허출원제10/368045호에 개시되는 바와 같이 인간 포피 지지세포층 상에서 배양하는 것이 가능하다. 포피 유래의 지지세포층은 인간 ES세포를 배양하기 위해 적합한 완전한 동물 비존재의 환경으로 구성된다. 또 포피 세포는 그 유도 이후, 42회의 계대배양의 길이에 걸쳐 배양상태로 유지하는 것이 가능하고, 그 결과 ES세포에는 비교적 일정한 환경이 제공된다. 이들 조건 하에서는 인간 ES세포는 대체 프로토콜(예를 들면, MEF)를 이용하여 성장된 세포와 기능적으로 상이한 것이 발견되었다. 분화후, ES세포는 인비트로 에서는 배의 3개의 배엽의 전체에 관련하는 유전자를 발현하고, 인비보에서는 3개의 배엽의 전체로부터 발생하는 조직으로 구성되는 기형종을 형성했다. 또 인간 난관 상피세포 또는 인간 배아섬유아세포와 달리, 포피 지지세포층에서 배양된 인간 ES세포는 적어도 87회의 계대배양에 걸쳐 다능성 의 미분화 상태로 배양에서 유지되었다. 그러나, 포피 세포는 장기간(즉, 42회의 계대배양)에 걸쳐 배양상태에 유지하는 것이 가능하지만, 포피 배양 시스템은 별개의 배취 사이에서의 차이로 인해 양호하게 규정되어있지 않다. 또 인간 지지세포층에 기초한 배양 시스템에서는 지지세포층 및 hES세포의 양자의 동시성장이 여전히 요구된다. 따라서, 다양한 지지세포 비함유 배양 시스템이 개발되고 있다.
Claims (152)
- 다능성 및 증식성의 미분화 상태로 유지될 수 있는, 지지세포 비함유의 인간 배아줄기세포주를 확립하는 방법으로서,지지세포를 함유하지 않으며, 매트릭스, 그리고 TGFβ1 및 bFGF가 보충된 조직배양배지를 포함하는 배양조건 하에서 상기 인간 배아줄기세포를 배양하여, 이로써, 지지세포 비함유의 인간 배아줄기세포주를 얻는 단계를 포함하는 것을 특징으로 하는, 인간배아줄기세포주를 확립하는 방법.
- 제1항에 있어서, 상기 배양조건 하에서의 상기 인간 배아줄기세포주로부터 세포를 클로닝하는 단계를 더 포함하는 것을 특징으로 하는 방법.
- 지지세포를 함유하지 않는 배양조건 하에서, 다능성 및 증식성의 미분화 상태로 인간 배아줄기세포주를 확대하는 방법으로서, 매트릭스, 및 TGFβ1과 bFGF가 보충된 조직배양배지 상에서 인간 배아줄기세포주의 세포를 배양하고, 이로써, 상기 인간 배아줄기세포주의 세포를 다능성 및 증식성의 미분화 상태로 유지하는 단계를 포함하는 것을 특징으로 하는 방법.
- 다능성 및 증식성의 미분화 상태로 유지될 수 있는, 이종 병원균 비함유 (xeno-free), 지지세포 비함유(feeder cell-free)의 특정종의 배아줄기세포주를 확립하는 방법으로서,지지세포가 없고 이종 병원균 (xeno pathogens)을 함유하지 않으며, 및 특정의 종 유래의 매트릭스, 그리고 TGFβ1 와 bFGF를 포함하는 조직배양배지를 포함하는 배양조건 하에서 배아줄기세포를 배양하고, 이로써, 이종 병원균 비함유 및 지지세포 비함유의, 특정 종의 배아줄기세포주를 얻는 단계를 포함하는 것을 특징으로 하는 방법.

- 제34항에 있어서, 상기 제조는,지지세포 및 이종병원균을 함유하지 않고, 및 인간 유래의 피브로넥틴 매트릭스와, TGFβ1과 bFGF가 보충된 조직배양배지를 포함하는 배양조건 하에서 인간 배아줄기세포를 배양하고, 이것에 의해 인간 배아줄기세포 배양물을 만드는 단계를 포함하는 것을 특징으로 하는 이종병원균 및 지지세포 미함유 인간 배아줄기세포 배양물.
- 제35항에 있어서, 상기 배양배지는 LIF를 더 포함하는 것을 특징으로 하는 이종병원균 및 지지세포 미함유 인간 배아줄기세포 배양물.
- TGFβ1과 bFGF를 함유하는 것으로서, 상기 TGFβ1 는 0.06-0.24 ng/ml의 농도범위로 제공되며 및 상기 bFGF는 적어도 2 ng/ml의 농도로 제공되는 것을 특징으로 하는 조직배양배지.
